针刺“百会”透“曲鬓”对JNK通路抑制后脑出血大鼠脑组织p38MAPK的影响

摘　　要：目的用针刺"百会"透"曲鬓"的方法治疗急性期脑出血模型大鼠,观察JNK通路被抑制后,p38MAPK的表达变化,探讨头针对出血后的脑组织是否起到保护作用。方法选用雄性SD大鼠120只随机分为假手术组、模型组、针刺组(模型+针刺组)、抑制剂组(模型+SP600125);选取自体血注入尾壳核方法造模,确定造模成功后给予针刺治疗,采取针刺方法为百会穴透曲鬓穴。在6 h、3 d、7 d 3个时间点取材,用TUNEL法检测各组大鼠脑出血半暗带区细胞凋亡情况;用Western blotting法观察脑组织内p38MAPK的蛋白表达。结果 TUNEL法:模型组,针刺组和抑制剂组与假手术组比较,差异有统计学意义(P <0.05)。针刺组与模型组比较在3、7 d有统计学意义(P <0.05)。针刺组与抑制剂组比较在3、7 d有统计学意义(P <0.05),抑制剂组与模型组比较各时间点均有显著差异(P <0.01)。Western blotting法:模型组,针刺组和抑制剂组与假手术组比较,差异有统计学意义(P <0.05)。针刺组与模型组比较,在3、7 d两个时间点表达比模型组降低明显,具有统计学意义(P <0.05)。针刺组与抑制剂组比较在3、7 d有统计学意义(P <0.05),抑制剂组与模型组比较在7 d有统计学意义(P <0.05)。结论针刺"百会"透"曲鬓"对大鼠脑出血后继发性脑损伤的神经细胞凋亡有抑制作用,并能降低p38MAPK的表达,起到了脑保护作用。Objective:To observe the changes of p38MAPK expression after JNK pathway was inhibited,and to detect whether head acupuncture has protective effect on brain tissue after hemorrhage.Methods:120 male SD rats were randomly divided into 4 groups:sham group,the model group,acupuncture group and inhibitor group.Autologous blood was injected into the caudate putamen nucleus to establish the model.Acupuncture was given after the success of the model establishment.Acupuncture was adopted at Baihui point and Qubin point.Samples were taken at the 6th hours,the 3rd days and the 7th days.TUNEL method was used to detect apoptotic cells in the penumbra of cerebral hemorrhage in rats of each group.The expression of p38MAPK protein in brain tissue was observed by Western blotting.Results:TUNEL method:There was significant difference among groups(P<0.05).There was significant difference on the 3rd day and the 7th day between the model group and the acupunc ture group,as well as between the acupuncture group and the inhibitor group(P<0.05).There were significant differences between the inhibitor group and the model group at different time points(P<0.01).Western blotting:There was significant difference among groups(P<0.05).Compared with the model group,the expression of acu puncture group was significantly lower on the 3rd day and the 7th day(P<0.05).There was statistical significance between inhibitor group and the model group on the 7th day(P<0.05).Conclusion:Acupuncture penetrating Baihui and Qubin can inhibit the apoptosis of neurons and decrease the expression of p38MAPK after cerebral hemorrhage in rats,with a protective effect on the brain.
