溃结宁膏穴位敷贴对溃疡性结肠炎大鼠结肠组织闭合蛋白影响的实验研究

摘　　要：目的观察溃结宁膏穴位敷贴对溃疡性结肠炎大鼠结肠黏膜的作用并探讨其作用机制。方法选用SD大鼠40只,随机分为空白组、模型组、敷贴组、SASP组,并建立溃疡性结肠炎大鼠模型,造模结束后,敷贴组给予溃结宁膏穴位敷贴,治疗结束后,观察大鼠宏观表征改变,计算疾病活动指数(DAI);Western blotting法检测大鼠结肠组织Claudin-1、Claudin-2、Claudin-8蛋白的表达。结果 1)治疗结束后,与模型组比较,敷贴组和SASP组的大鼠宏观表征改变有一定程度的改善(P <0.01);2)与空白组比较,其余3组的Claudin-2蛋白表达上升(P <0.01),Claudin-1、Claudin-8等蛋白的表达下降(P <0.01);与模型组比较,敷贴组和SASP组上述蛋白指标均显著改善(P <0.05);与SASP组比较,敷贴组各项指标差异不明显(P> 0.05)。结论溃结宁膏穴位敷贴可以一定程度上降低实验大鼠Claudin-2蛋白的水平,同时使得Claudin-1、Claudin-8的蛋白表达上升,从而达到减轻肠道炎症反应,促进结肠黏膜愈合,重建肠黏膜机械屏障的完整性的目的。Objective:To observe the effect of KuiJieNing Ointment acupoint application on colonic mucosa of rats with ulcerative colitis,and to explore its specific mechanism of action on ulcerative colitis.Methods:40 SD rats were randomly divided into the blank group,the model group,the application group and SASP group,and rat models of ulcerative colitis were established.After the model was established,the application group was given acu puncture point application.After treatment,the macroscopic characterization changes were observed and DAI was calculated.The expression of Claudin-1,Claudin-2 and Claudin-8 protein in rat colon tissue was detected by Western blotting.Results:After treatment,compared with the model group,the macroscopical changes of rats in the application group and SASP group were improved to some extent(P<0.01).Compared with the blank group,Claudin-2 protein expression increased in the other three groups(P<0.01),while Claudin-1 and Claudin-8 protein expression decreased(P<0.01).Compared with the model group,the abovementioned protein indexes in the application group and SASP group changed correspondingly(P<0.05);there was no significant difference in each index between the application group and SASP group(P>0.05).Conclusion:Acupoint application with KuiJieNing Ointment can reduce the level of Claudin-2 protein in experimental rats to some extent,and increase the protein expression of Claudin-1 and Claudin-8,so as to alleviate intestinal inflammation,promote the healing of colonic mucosa and reconstruct the integrity of intestinal mucosal mechanical barrier.
