摘 要:目的验证沙苑子总黄酮成分是否具有促进骨髓间充质干细胞(BMSCs)向神经细胞分化的作用。方法培养获得SD大鼠的BMSCs并鉴定。选择沙苑子总黄酮并同单唾液酸四己糖神经节苷脂(GM1)比较,观察其刺激体外培养SD大鼠的BMSCs后,细胞形态学变化,免疫细胞化学检测神经元特异性烯醇化酶(NSE)的表达,及Western blotting检测巢蛋白(Nestin)的表达。结果沙苑子总黄酮刺激后BMSCs的细胞形态出现有类似神经元细胞样改变;免疫细胞化学检测到沙苑子总黄酮刺激后的部分细胞有NSE的表达;Western blotting检测到沙苑子总黄酮刺激后的细胞样本有Nestin的表达。结论沙苑子总黄酮能促进体外培养BMSCs向神经细胞分化的作用。Objective:To verify whether the total flavonoids of Astragalus can promote the differentiation of BMSCs into neurons.Methods:BMSCs of SD rats were cultured and identified.Total flavonoids of Astragalus were selected and compared with GM1 to observe the morphological changes of BMSCs cultured in SD rats stimulated by GM1.The expression of NSE was detected by immunocytochemistry.Nestin expression was detected by Western blotting.Results:The morphology of BMSCs stimulated by total flavonoids of Astragalus showed neuron-like changes.NSE expression was detected in some cells stimulated by total flavonoids of Astragalus mongolica by immunocytochemistry,and Nestin expression was detected by Western blotting.Conclusion:The total flavonoids of Astragalus can promote the differentiation of BMSCs into neurons in vitro.
