摘 要:目的分析慢性肺阻塞疾病(COPD)患者滋养层细胞表面抗原2(TROP2)表达及临床意义。方法选取2017年8月~2018年8月期间在我院接受治疗肺部手术COPD患者和非COPD患者各52例,分别作为COPD组和对照组,在获取患者同意基础下取肺部组织。肺部组织TROP2表达情况分析,比较两组肺部组织TROP2表达以及肺功能,免疫功能,COPD患者TROP2与肺功能、免疫功能相关性。结果TROP2免疫染色结果显示COPD患者肺部阳性表达TROP2显示为棕褐色,而对照组TROP2表达量较少,对基底细胞标记物Cytokeration以及TROP2双免疫荧光染色结果显示,TROP2表达于气道基底细胞;COPD组患者TROP2相对表达量显著高于对照组(P<0.05),FEV 1和FEV 1/FVC显著低于对照组(P<0.05);COPD组患者CD+4、CD+8以及CD+4/CD8均低于对照组,差异具有统计学意义(P<0.05);COPD患者TROP2与FEV 1和FEV 1/FVC等肺功能指标以及CD+4、CD+8等免疫功能指标负相关(P<0.05),而与CD+4/CD+8无相关性(P>0.05)。结论COPD患者气道基底细胞大量表达TROP2,且其与患者肺功能以及免疫功能关系密切。Objective To analyze the expression of trophoblast cell-surface antigen 2(TROP2)in patients with chronic obstructive pulmonary disease(COPD)and its clinical significance.Methods 52 patients with COPD and 52 patients without COPD who underwent pulmonary surgery in the hospital from August 2017 to August 2018 were included in the COPD group and the control group.The lung tissues of patients were collected on the basis of obtaining their consent.The expression of TROP2 in lung tissues was analyzed.The expression of TROP2 in lung tissues,lung function and immune function were compared between the two groups.The correlation of TROP with lung function and immune function in COPD patients was analyzed.Results The results of TROP2 immunostaining showed that the positive expression of TROP2 in lung of COPD patients was brown,while the expression of TROP2 in the control group was less.The results of double immunofluorescence staining of basal cell markers Cytokeration and TROP2 showed that TROP2 was expressed in airway basal cells.The relative expression of TROP2 in the COPD group was significantly higher than that in the control group(P<0.05),while FEV 1 and FEV 1/FVC were significantly lower than those in the control group(P<0.05).The levels of CD+4,CD+8 and CD+4/CD+8 were lower in the COPD group were lower than those in the control group.(P<0.05).TROP was negatively correlated with lung function indexes such as FEV 1 and FEV 1/FVC,and immune function indexes such as CD+4 and CD+8 in COPD patients(P<0.05),while not correlated with CD+4/CD+8(P>0.05).Conclusion TROP2 is highly expressed in airway basal cells of COPD patients,and it is closely related to lung function and immune function.
