摘  要:目的:研究白芷提取物对大肠癌荷瘤裸鼠的抗肿瘤及其免疫功能调节作用,探讨其抗肿瘤的免疫学机制。方法:采用大肠癌细胞株(CT26)BALB/c nude荷瘤裸鼠模型,白芷提取组分(AD-2P)联合氟尿嘧啶(5-FU)、亚叶酸(leucovorin,LV)给药14天后,称重计算瘤重、瘤体积、脾脏、肝脏和胸腺指数等;用酶联免疫吸附法(ELASA)检测RAW264.7细胞株下使用AD-2P后的NO、TNF-α分泌水平。采用Western blot法检测白芷提取物对RAW264.7细胞株MAPKs通路下ERK1/2、JNK和P38等蛋白的表达。结果:AD-2P协同5-Fu和LV治疗组能抑制大肠癌荷瘤裸鼠肿瘤的生长(P<0.05);AD-2P可增加RAW264.7细胞NO与TNF-α分泌量(P<0.05),并呈现剂量依赖关系;Western blot实验中,在不同浓度的AD-2P作用后,RAW264.7细胞的MAPK通路ERK1/2、JNK与P38的磷酸化活化产物P-ERK1/2、P-JNK及P-P38的蛋白表达量增加,并呈一定的时间和梯度依赖。结论:AD-2P可能通过增加NO和TNF-α产物,激活MAPK通路的ERK1/2、JNK和P38蛋白表达发挥免疫调节作用,进而抑制大肠癌瘤体的增长,此研究为开发新的抗肿瘤药物提供了可能。Objective:To investigate the anti-tumor and immunomodulatory effects of Angelica dahurica extract(AD-2P)on colorectal cancer in CT26 tumor-bearing mice,and to explore the immunomodulatory mechanism against colorectal cancer.Methods:The CT26 tumor-bearing BALB/c mice model was adopted.After 14-day administration of AD-2P combined with 5-FU and leucovorin(LV),the body weight,tumor volumes,and indexes of spleen,liver and thymus were measured.The secretion levels of NO and TNF-αin RAW264.7 cell line after AD-2P treatment was detected by ELASA method.Western blot method was used to detect the proteins expressions of ERK1/2,JNK and P38 of MAPKs pathway in RAW264.7 cell line.Results:AD-2P combined with 5-Fu and LV significantly inhibited the growth of colorectal cancer in nude mice(P<0.05).AD-2P could greatly increase the NO and TNF-αsecretions in RAW264.7 cells(P<0.05),which showed a dose-dependent relationship.In Western blot experiment,the phosphorylated activation products expressions(P-ERK1/2,P-JNK and P-P38)of MAPK pathway were increased in RAW264.7 cell line by a time-dependent and dose-dependent manner,after different concentrations of AD-2P intervention.Conclusion:AD-2P can inhibit the growth of colorectal cancer by increasing NO and TNF-α,activating the protein expressions of ERK1/2,JNK and P38 of the MAPK pathway,to play the role of immune regulation.It provided the reference for further research and development of new anti-cancer drugs.
