摘 要:目的探讨多模态MRI评估恒河猴慢性酒精暴露肝损伤的可行性。方法利用7只健康雄性恒河猴建立慢性酒精暴露肝损伤模型,于建模前(P0期)及酒精诱导期结束后持续喂养1月(P1期)、3月(P2期)及6月(P3期),并于各维持期末行相同的多模态MRI扫描,采用单因素方差分析方法分析不同时间点多模态MRI参数间的差异,进一步分析采用SNK-q检验法,用Pearson相关性分析的方法分析多模态MRI参数和血液学指标之间的相关性。结果建模成功。表观扩散系数(ADC)值及真性扩散(D)值在部分时间点之间差异有统计学意义,ADC值及D值与三酰甘油指标呈显著负相关;假性扩散(D*)及灌注分数(f)值在各时间点之间差异无统计学意义;D*及f值与血液学指标之间均无显著相关;脂肪分数(FF):在部分时间点之间差异有统计学意义;FF与三酰甘油、AST指标呈显著正相关;ADC值、D值与FF呈显著负相关。结论慢性酒精暴露会对肝脏造成损伤,利用多模态MRI可对其进行定量评价,非人灵长类动物疾病模型的建立对临床相关疾病研究具有重要参考价值。Objective To evaluate the liver injury model caused by chronic alcohol exposure in rhesus monkeys by multimodal MRI.Methods The liver injury models were caused by chronic alcohol exposure with seven rhesus monkeys.The same multimodal MRI scans were performed before alcohol induction(P0)and continuous feeding for one month(P1),three months(P2),six months(P3)after alcohol induction.The comparison of multimodal magnetic resonance parameters at different time points were statistically analyzed with one-way ANOVA.The SNK-q test was used for further analysis.The correlation between multimodal magnetic resonance parameters and hematological parameters were statistically analyzed with Pearson correlation.Results The liver injury model caused by chronic alcohol exposure in rhesus monkeys was successfully established.There were significant differences in apparent diffusion coefficient(ADC)value and true diffusion(D)value between some time points.ADC value and D value were significantly negatively correlated with triglyceride indicators.Pseudodiffution(D*)value and perfusion fraction(f)value were not significantly different between all time points.D*value and f value were not significantly correlated with hematological indicators.Fat fraction:There were significant differences between some time points.Fat fraction was significantly positively correlated with triglyceride and AST indicators.ADC value and D value were significantly negatively correlated with fat fraction.Conclusion Chronic alcohol exposure can cause liver injury,which can be quantitatively evaluated by multimodal MRI.The establishment of non-human primate disease model has important reference value for clinical research.
