摘  要:为探讨自噬在Cd胁迫小白菜生长中的作用,采用营养液培法研究了Cd胁迫下小白菜生长量、叶绿素、活性氧、MDA及抗氧化酶活性与自噬的关系。结果表明:随Cd^2+浓度增加,小白菜生长受到明显抑制,叶片褪绿,根系变短,并产生细胞自噬;叶绿素荧光参数Fm、Fv/Fm和Qp显著降低,而Qn、Y(NPQ)显著升高,SPAD值则“先升后降”;H2O2、O2^-·均呈上升趋势,差异显著,其中O2^-·增幅较H2O2大;SOD、CAT、APX及总抗坏血酸值为“先升后降”,而GSH-Px、ASA则持续上升,差异显著;根、叶中MDA含量升高,且叶比根过氧化损伤更为严重。由此,Cd应答可激活小白菜发生自噬;随Cd^2+浓度升高,ROS累积诱导自噬小体形成,然而自噬发生不足或过量都直接导致植株DNA损伤和程序性细胞死亡。In order to explore the effects of autophagy on the growth of Chinese cabbage under Cd stress,the relationship between growth,chlorophyll,reactive oxygen species,MDA,antioxidant enzyme activity and autophagy under Cd stress in Chinese cabbage was studied by a hydroponic experiment.The results showed that with the increase of Cd2+concentration,the growth of pakchoi was significantly inhibited,showing leaf chlorosis,root shortening and cell autophagy;the chlorophyll fluorescence parameters Fm,Fv/Fm and Qp decreased significantly,while Qn and Y(NPQ)increased significantly,and SPAD values increased first and then decreased;the increase of H2O2 and O2^-·was significant,and the increase of O2^-·was more dramatic than that of H2O2;SOD,CAT,APX and total antiserum acid value increased first and then decreased,while GSH-Px and ASA increased continuously with significant difference;the content of MDA in roots and leaves increased,and the damage to leaves was more serious than that to roots.In conclusion,Cd response is the cause of activation of autophagy in Chinese cabbage.With the increase of Cd2+concentration,ROS accumulation induced autophagosome formation.However,inadequate or excessive autophagy could directly lead to DNA damage and programmed cell death in plants.
