摘  要:目的通过TUNEL法检测白藜芦醇(RES)对佐剂性关节炎(AA)大鼠滑膜细胞的凋亡影响,探讨RES对AA大鼠应用效果。方法0.1mL弗氏完全佐剂注射SD大鼠左足趾皮下诱导关节炎复制AA模型,24d后,取非致炎侧踝关节小块滑膜组织分离、培养滑膜细胞,将传代后的滑膜细胞加入不同浓度的RES溶液,利用TUNEL法观察滑膜细胞凋亡情况。结果TUNEL法观察到加入高浓度(20、50μmol/L)RES后TUNEL阳性细胞数明显增多。结论RES能促进AA大鼠滑膜细胞的凋亡。Objective To investigate the effect of resveratrol (RES) on apoptosis of synovial cells in adjuvant arthritis (AA) rats by TUNEL method. Methods 0.1 mL Freund s complete adjuvant was injected into the left toe of SD rats to induce arthritis and replicate AA model. After 24 days, non-inflammation small synovial tissues of lateral ankle joint were isolated and synovial cells were cultured. Synovial cells after passage were added to different concentrations of RES solution, and TUNEL staining was used to observe the apoptosis of synovial cells. Results TUNEL staining showed that the number of TUNEL-positive cells increased significantly after the addition of high concentration RES (20 and 50 micromol/L). Conclusion RES could promote the apoptosis of synovial cells in AA rats.
