摘 要:目的探讨与评价过表达Bestrophin 3对皮质类固醇诱导的股骨头坏死(osteonecrosis of the femoral head,ONFH)保护作用及其机制。方法根据纳入标准和排除标准选取本院于2017年2月至2017年12月期间就诊的ONFH患者术后股骨头骨组织50例作为观察组,股骨颈骨折患者自愿捐赠的股骨头骨组织50例作为对照组,采集两组标本的骨髓基质细胞,使用PCR方法检测骨髓基质细胞Bestrophin 3的表达水平。利用分子生物学手段构建Bestrophin 3不同表达水平的细胞株,通过MTT检测细胞生存率方法比较不同细胞株中细胞生存率。结果观察组中的股骨头骨组织Bestrophin 3相对表达量高于对照组,差异有统计学意义(P<0.05)。高表达Bestrophin 3的细胞株细胞生存率比较高,差异有统计学意义(P<0.05),且细胞生存率与Bestrophin 3表达量呈正相关(P<0.01)。结论Bestrophin 3在皮质类固醇性股骨头坏死骨组织中的呈现高表达状况,Bestrophin 3表达量与细胞生存率呈正相关,高表达Bestrophin 3对细胞具有保护作用。Objective To explore and evaluate the protective effect of overexpressing Bestrophin 3 on cor ticosteroid induced femoral head necrosis and its mechanism.Method In accordance with the inclusion criteria and exclusion criteria,femoral head tissues from 50 patients after cortical steroid induced femoral head necrosis from February to December 2017 in our hospital were selected as observation group and 50 cases of femoral skull tissue donated by patients with femoral neck fractures as control group.Two groups of specimens of bone marrow stromal cells were collected,and the expression level of bone stromal cells Bestrophin 3 was detected by PCR.The cell lines with different expression levels of Bestrophin 3 were constructed by molecular biology methods.Cell survival rate of different cell lines was compared by MTT assay.Result The relative expression of Bestrophin 3 in femoral skull tissue in the observation group was higher than that in the control group,and the difference was statistically significant(P<0.05).Cell survival rate of cells with high expression of Bestrophin 3 was higher(P<0.05),which was statistically significant and that was positively correlated with the expression of Bestrophin 3(P<0.01).Conclusion Bestrophin 3 is highly expressed in steroid induced osteonecrosis of femoral head.The expression of Bestrophin 3 is positively correlated with cell survival rate and the cells are protected by the high expression of Bestrophin 3.
