摘  要:目的:探讨失荅刺知丸及拆方对脑缺血再灌注模型大鼠脑组织JNK表达的影响。方法:将204只SD雄性大鼠按随机数字表法分为正常组、假手术组、模型组及干预组(包括失荅剌知丸组,失荅剌知丸拆方Ⅰ组、拆方Ⅱ组、拆方Ⅲ组),其中模型组及各干预组按时间点不同分为1 d组、3 d组和7 d组,并予MACO线栓法制备脑缺血再灌注大鼠模型,同时干预组于每日固定时间灌胃,同时观察大鼠神经功能症状情况,以大鼠神经功能Garcia JH评分并记录,治疗后于相应时间取材。运用Western Blot法检测海马组织中p-JNK蛋白的含量,以p-JNK/GAPDH表示蛋白表达相对水平。结果:失荅剌知丸及各拆方组中的p-JNK蛋白表达量均有降低,且与灌药时间成正比(P<0.05);失荅剌知丸原方较各拆方组蛋白表达均低,其中拆方Ⅰ组最高,拆方Ⅱ组最低,拆方Ⅲ组次之(P<0.05)。神经功能评分结论与p-JNK蛋白表达结果一致。结论:失荅剌知丸原方及各拆方组均具有改善脑缺血后神经功能、抗神经细胞凋亡作用,而这可能与本实验所研究的JNK蛋白下调有关;其中,除原方外,又以拆方Ⅱ组作用效果最好,考虑该组可能在失荅剌知丸原方中发挥君药、或君药加臣药的主要作用。Objective To investigate the effects of the Shidalazhiwan and its dismantling prescriptions on the expression of JNK in the brain tissue of rats with cerebral ischemia-reperfusion.Methods 204 male Sprague-Dawley rats were randomly divided into normal group,sham operation group,model group and intervention group(including the Shidalazhiwan group,the Dismantling prescription group I,the Dismantling prescription group II,the Dismantling prescription group III),in which the model group and the intervention group were divided into 1 d group,3 d group and 7 d group according to the time point,and the rat model of cerebral ischemia-reperfusion was prepared by MACO suture method,and the intervention group was fixed daily.At the same time,the neurological symptoms of the rats were observed,and the neurological function Garcia JH score will be scored and recorded.After the treatment,the materials were taken at the corresponding time.The content of p-JNK protein in hippocampus was detected by Western Blot,and the relative level of protein expression was expressed by p-JNK/GAPDH.Results The expression levels of p-JNK protein in the Shidalazhiwan and the disassembled prescription groups were all decreased,and it was proportional to the time of drug injection(P<0.05).The original expression of Shidalazhiwan was lower than that of the disassembled prescriptions.The disassembled group I was the highest,the disassembled group II was the lowest,and the disassembled group III was the second(P<0.05).The neurological function score was consistent with the p-JNK protein expression.Conclusion The original Shidalazhiwan and the disintegration prescriptions have the effects of improving neurological function and anti-neuronal apoptosis after cerebral ischemia,which may be related to the down-regulation of JNK protein studied in this experiment.The effect of dismantling prescription II is the best.Considering that this group may play the main role of the drug in the original prescription of the sputum,or the drug of the genus.
