摘  要:以蓝莓品种‘北陆’‘斯巴坦’的组培苗叶片为外植体,研究其离体培养技术。结果表明:在含有1.5 mg/L ZT、0.3 mg/L IAA、30 g/L庶糖、4.5 g/L琼脂、pH值4.5的改良WPM培养基上进行离体叶片再生培养,最早37.7 d观察到再生植株。60 d后‘北陆’再生率最高达到93.5%,‘斯巴坦’再生率最高达到89.8%,‘北陆’再生效果优于‘斯巴坦’。‘北陆’叶片再生的适宜pH值为5.0,斯巴坦为4.5。pH值过高或过低再生率降低,愈伤组织形成少或无,叶片发黄、最后枯萎。不同叶片处理方式和暗培养对蓝莓叶片再生影响均不大。With‘Northland’and‘Spartan’,the somaclone of temple leaves as the explants,the containing ZT 1.5 mg/L,IAA 0.3 mg/L,30 g/L total sugar,4.5 g/L of AGAR,pH value to 4.5 improved cultivated in vitro on leaf regeneration WPM medium.the first 37.7 d visible regeneration plant was observed,after 60 d investigation regeneration rate is highest,‘Northland’regeneration rate of up to 93.5%,‘Spartan’regeneration rate of up to 89.8%,‘Northland’regeneration effect is better than‘Spartan’.The pH value of regeneration culture of leaves in vitro of‘Northland’is 5.0,‘Spartan’is 4.5.When the pH value is too high or too low,regeneration rate lower,callus formation,less or no,leaf yellow,eventually wither.Different leaf treatments and dark culture had little effect on leaf regeneration of blueberry.
