基于"肝藏血"理论采用"养血柔肝"法的中药复方对肝纤维化细胞因子的影响

摘　　要：目的:研究养血柔肝丸对大鼠肝纤维化细胞因子的影响。方法:选用雌雄各半大鼠,随机分为正常对照组、模型组、养血柔肝丸6. 25 g/kg、12. 5 g/kg和25. 0 g/kg组、扶正化瘀胶囊0. 75 g/kg组。采用50% CCl4(1 ml/kg)复制肝纤维化模型。按组别分别给药治疗,治疗结束后,测定血清白介素-1(IL-1)、白介素-6(IL-6)、转化生长因子β(TGF-β1)、肿瘤坏死因子α(TNF-α),取肝制备10%肝匀浆,测定其丙二醛(MDA)、超氧化物歧化酶(SOD)、羟脯氨酸(HYP)及基质金属蛋白酶抑制因子1(TIMP-1)水平,并做肝脏病理组织学检查。结果:肝纤维化模型大鼠造模后,血清IL-1、IL-6、TNF-α及TGF-β1显著升高;大鼠肝匀浆MDA活性显著升高、SOD活性显著下降;TIMP-1和HYP的含量明显升高,说明肝纤维化模型成立。给予养血柔肝丸治疗后,大鼠血清中IL-1、IL-6、TNF-α和TGE-β1含量显著降低;肝匀浆MDA、TIMP-1及HYP含量显著降低,SOD含量显著升高;肝脏病理损伤明显改善。结论:养血柔肝丸对大鼠四氯化碳肝纤维化因子有明显的影响,起效剂量为6. 25 g/kg。Objective: To study the effect of Yangxue Rougan Pill on cytokines of liver fibrosis in rats. Methods: Rats were randomly divided into six groups: the control group,the model group,Yangxue Rougan Pill groups (25. 0 g/kg,12. 5 g/kg and 6. 25 g/kg),Fuzheng Huayu Capsule group (0. 75 g/kg). The liver fibrosis model was replicated by 50% CCl4 (1 ml/kg). After the administration,the levels of serum IL-1,IL-6,TGF-β1,TNF-αwere measured. Liver tissues were collected and 10% liver homogenate was prepared to determine the levels of MDA、SOD、HYP and TIMP-1,and histopathological examinations were taken. Results: After modeled,serum levels of IL-1,IL-6,TNF-α and TGF-β1 were significantly increased,MDA content was significantly increased and SOD activity was significantly decreased in rat liver homogenate,contents of TIMP-1 and HYP were significantly increased,which showed the liver fibrosis model was established. After treatment with Yangxue Rougan Pill,levels of serum cytokines IL-1,IL-6,TNF-α and TGE-β1 were significantly decreased,levels of MDA,TIMP-1 and HYP in liver homogenate were significantly decreased,SOD level was significantly elevated,and histopathologic damages were improved.Conclusion: Yangxue Ruogan pill has an obvious effect on rat carbon tetrachloride hepatic fibrosis cytokines,and the effective dose is 6.25 g/kg.
