儿童支原体肺炎患者外周血单核细胞中TLRs的表达及其对NF-kB/IkBα信号通路的影响

摘　　要：目的探讨PBMC中TLRs在儿童支原体肺炎中的表达及对NF-κB/IκBα信号通路的影响。方法选择本院2014年7月-2016年9月科室收治的儿童支原体肺炎患者22例为支原体肺炎组,非支原体肺炎儿童25例为非支原体肺炎组,同期健康体检儿童25例为对照组。对3组PBMC中TLRs表达进行检测并探讨后者对NF-κB/IκBα信号通路的影响。结果非支原体肺炎组、支原体肺炎组TLR1mRNA、TLR2mRNA、TLR4mRNA、TLR7mRNA高于对照组(P<0.05);3组血清TNF-α、IL-1β、IL-18相比较,支原体肺炎组>非支原体肺炎组>对照组(P<0.05);3组IκBαmRNA、p65和p-p65的蛋白表达水平差异明显,差异均有统计学意义(P<0.05)。结论儿童支原体肺炎患者PBMC中TLRs的表达存在部分异常改变且与炎症反应NF-kB/IkBα信号通路激活有关。Objective To investigate the expression of TLRs in childhood Mycoplasma pneumoniae and its effect on NF-κB/IκBαsignaling pathway in PBMC.Methods Twenty-two children with mycoplasmal pneumonia admitted to our department from July 2014 to September 2016 were enrolled in the mycoplasma pneumonia group.25 children with non-mycoplasmal pneumonia were in the non-mycoplasmal pneumonia group,and 25 healthy children in the same period were collected as the control group.The expression of TLRs in three groups of PBMC was detected and the effect of the latter on NF-κB/IκBαsignaling pathway was investigated.Results The TLR1 mRNA,TLR2 mRNA,TLR4 mRNA and TLR7 mRNA in non-mycoplasma pneumonia group and mycoplasma pneumonia group were higher than those in the control group(P<0.05).Serum TNF-α,IL-1βand IL-18 were compared among the three groups,showing that Mycoplasma pneumonia group>non-mycoplasmal pneumonia Group>control group(P<0.05);the protein expression levels of IκBαmRNA,p65 and p-p65 in the three groups were significantly different,with the differences statistically significant(P<0.05).Conclusion The expression of TLRs in PBMC of children with Mycoplasmal pneumonia is partially abnormal and related to the activation of NF-κB/IκBαsignaling pathway.
