摘 要:目的本文研究黄腐醇对NCI-H460细胞凋亡的作用机制研究。方法常规培养NCI-H460肺癌细胞,采用MTT法检测黄腐醇对NCI-H460细胞生长的影响;通过细胞集落实验和流式细胞术检测黄腐醇对NCI-H460细胞集落形成和细胞凋亡影响;采用Western Blotting法检测黄腐醇对NCI-H460细胞内Bcl-2和Bax蛋白的影响。结果黄腐醇能够有效抑制NCI-H460的生长,其IC 50值为6.50μmol·L^-1,并能浓度依赖性的抑制NCI-H460细胞集落的形成及促进细胞凋亡;同时,黄腐醇呈浓度依赖性地促进促凋亡蛋白Bax的表达,和抑制抑凋亡蛋白Bcl-2的表达。结论黄腐醇对NCI-H460细胞的生长和凋亡作用呈浓度依赖性,并通过激活促凋亡蛋白Bax和抑制抑凋亡蛋白Bcl-2的表达促进细胞凋亡。OBJECTIVE To investigate the mechanism of action of xanthohumol on apoptosis of NCI-H460 cells.METHODS The NCI-H460 lung cancer cells were cultured by conventional method,the effect of xanthohumol on the growth of NCI-H460 cells was detected by MTT assay;The effects of xanthohumol on colony formation and apoptosis of NCI-H460 cells were detected by cell colony assay and flow cytometry,respectively;Western Blotting was used to detect the expression of Bax and Bcl-2 proteins after treated with xanthohumol.RESULTS The results showed that xanthohumol inhibited effectively NCI-H460 lung cancer,with the IC 50 value on NCI-H460 cancer cells as 6.50μmol·L^-1.Xanthohumol restrained the NCI-H460 colony formation and promoted NCI-H460 apoptosis in a concentration-dependent pattern.Besides,xanthohumol promoted the expression of the pro-apoptotic proteins Bax,and inhibited the expression of the anti-apoptotic protein Bcl-2 in a concentration-dependent manner.CONCLUSION The effect of xanthohumol on the growth and apoptosis of NCI-H460 cells in a concentration-dependent manner,and xanthohumol promoted pro-apoptotic protein Bax and inhibited the expression of the apoptotic protein Bcl-2 to induce the apoptosis of NCI-H460 cancer cells.
