摘  要:以西藏红景天为研究材料,利用DNA条形码(核糖体内转录间隔区ITS以及两对叶绿体DNA序列rbcL和trnS-G),对所收集的44个样品进行克隆与测序,并结合NCBI中已报道的22种红景天ITS,rbcL,trnS-G序列进行分析.利用Mega5.0软件基于DNA条形码序列分析上述样本的遗传距离结果显示,ITS和trnS-G序列在红景天属植物种间的差异明显,rbcL序列的差异较小.所构建的N-J进化树结果显示ITS对红景天物种的识别能力优于rbcL和trnS-G,但任何单一的DNA条形码均无法成功辨识全部的红景天属植物.其中ITS序列可以清晰的分辨出大花红景天与圣地红景天,并且不同地理居群间的样本差异较小,rbcL+trnS-G组合DNA条形码可以清晰分辨出长鞭红景天.本研究对西藏红景天属植物进行了遗传多样性分析,为其开发与利用提供指导.Using Tibet Rhodiola species as the research objects, three DNA barcode sequences include ribosomal transcribed spacer(ITS) and chloroplast DNA sequences rbcL gene and trnS-G were amplified from 44 Rhodiola samples of different geographical populations around Tibet. These sequences with the other 22 Rhodiola species sequences which had been reported in NCBI were analysis by Mega 5.0 software.The results showed that the ITS and trnS-G sequences genetic distance between all samples were significantly different, and the rbcL sequences were small. According to the N-J phylogenetic tree, ITS sequence was better than rbcL and trnS-G to investigate the Rhodiola species. And none single DNA barcode could successfully identify all Rhodiola plants. In addition, ITS sequence can clearly distinguish the Rhodiola crenualta and Rhodiola sacra with the small differences between the samples of different geographical population. By rbcL+trnS-G combined DNA barcodes, the Rhodiola fastigiata could be clearly distinguished. This paper carried out genetic diversity analysis of Rhodiola species in Tibet and provided guidance for their development and utilization.
