摘  要:目的研究肉苁蓉多糖、松果菊苷对快速老化模型小鼠SAMP8行为学及肠道菌群的影响。方法利用85%乙醇提取、D101大孔树脂分离结合制备液相分离制备松果菊苷;通过水提取、乙醇沉淀的方法利用药渣制备肉苁蓉多糖,然后对SAMP8小鼠每天进行灌胃,连续31 d,利用Morris水迷宫实验进行学习记忆能力测试;无菌条件下采集各组小鼠粪便,提取基因组DNA,对样本的16S rDNA基因V4区进行PCR扩增,采用Illumina HiSeq测序平台进行高通量测序,并对样本进行生物信息学分析。结果肉苁蓉多糖单独给药及与松果菊苷联合给药均能延长SAMP8小鼠生存率,改善小鼠生存质量。Morris水迷宫实验结果显示肉苁蓉对SAMP8小鼠学习记忆能力无显著影响。粪便肠道菌群α多样性和β多样性分析结果显示,与对照组相比,模型组菌群多样性升高,松果菊苷能提高主要拟杆菌、变形菌的数量,降低厚壁菌门数量,但是变化无统计学意义。结论肉苁蓉多糖单用及与松果菊苷联合使用能够提高SAMP8小鼠生存率且对SAMP8小鼠的肠道菌群紊乱具有改善和恢复作用。Objective To evaluate the efficacy of polysaccharides and echinacoside in the treatment of Alzheimers disease using senescence-accelerated prone mice(SMAP8). Methods The echinacoside was separated from the 85% ethanol extraction, by D101 types of macroporous resin and followed by preparative liquid chromatography. Then the residue extracted with water, precipitation with ethanol, dried by lyophilizer, and cistanche crude polysaccharide was obtained.SAMP8 mice were orally administered with echinacoside and crude polysaccharide with the method of single or combined administration for 31 d. Experiment Morris water maze was used to detect spatial learning and memory ability.Mouse feces were collected under sterile conditions and genomic DNA was extracted. High-throughput sequencing was carried out using illumina HiSeq sequencing platform, and bioinformatics was analyzed. Results Echinacoside and polysaccharides were administered single or combined both can prolong the survival rate and improve the quality of survival in mice. However, our results showed that cistanche chemical components had no use in learning and memory ability. Analysis of alpha and beta diversity of gut microbiota showed that the diversity of the model group increased compared to the control group. Echinacoside and combined group can increase the abundance of Bacteroides,Proteobacteria and reduced the abundance of Firmicutes, but no statistical significance. Conclusion Echinacoside and polysaccharides and the combination with can improve the survival rate of SAMP8 mice and restore gut microbiota disorders.
