摘 要:为了筛选烟草黑胫病抗性材料,本研究利用与黑胫病抗性基因紧密连锁的五个分子标记对中国烟草总公司陕西分公司提供的60份烟草育种亲本材料、65份杂交种材料和35份烟草育种高代材料的黑胫病抗性基因进行特异性分子标记检测,同步进行苗期温室抗黑胫病特性鉴定。结果表明:有9份、48份、7份、39份、51份和6份材料分别对烟草黑胫病表现为高抗、中抗、抗病、感病、中感和高感。在64份对黑胫病表现为抗性的材料中,含5个连锁标记的有42份,占比65.6%;供试材料中均含有黑胫病抗性基因连锁标记的有86份,占供试材料的53.8%,其中抗性材料42份,占比48.8%。相关性分析表明:烟草品种病情指数与抗性基因的5个紧密连锁标记的有无均极显著相关,含有抗性基因的材料病情指数极显著低于不含抗性基因材料的病情指数。在利用分子标记辅助选择育种过程中,不同类型材料选择的最优标记不同。本研究供试的三类材料中亲本材料可优先选择连锁标记PT20383、Scf30K;高代组合材料优先选择连锁标记INDel10617;不育系杂交材料可以选择连锁标记PT20383。本研究为烟草抗黑胫病改良中亲本选配、抗性基因的利用及杂交后代抗性基因的特异分子标记选择提供参考。In order to screen tobacco black shank disease resistant materials, 60 tobacco breeding parent materials,65 hybrid materials and 35 tobacco breeding high-grade materials was specifically detected by molecular markers closely linked to the black shank resistance gene, and the characterization of the resistance to black shank in the greenhouse was simultaneously carried out. The results were 9, 48, 7, 39, 51 and 6 materials showed high resistance, medium resistance, disease resistance, susceptible, moderate and high sensitivity to tobacco black shank. 42 materials contained 5 linkage markers in 64 anti-black rickets materials, accounting for 65.6%;86 of the tested materials contained resistance gene linked markers, accounting for 53.8% of the tested materials. Among them, 42 resistant materials accounted for 48.8%. Correlation analysis showed that the disease index of tobacco varieties was significantly correlated with the presence or absence of five closely linked markers of resistance gene.The disease index of materials containing resistance gene was significantly lower than that of gene-free material. In the process of using molecular marker-assisted selection breeding, the optimal markers for different types of material selection are different. In this study, the parental materials in the test materials were preferentially selected as linkage markers PT20383 and Scf30 K, and the high-generation composite materials were preferentially selected as linkage marker INDEL10617;the sterile hybridization materials were selected as linkage marker PT20383. To provide a data reference for the selection of parents in the improvement of tobacco resistance to black shank, the utilization of resistance genes and the selection of specific molecular markers for hybrid progeny resistance genes.
