摘　　要：本研究以新鲜西洋参根为外植体,研究其不定根诱导及发生特点。以MS为基础培养基,通过添加外源激素诱导不定根,观察外植体上不定根发生和增殖生长的特征。结果表明:西洋参不定根的发生过程包括外植体愈伤组织发生、愈伤组织胚性化、不定根分化和生长等四个阶段;在进行不定根继代时,带有愈伤组织的不定根生长速度更快,而且携带的愈伤组织在生长的同时,其表面也会继续分化出新不定根;另外,外植体体积对不定根诱导有很大影响,同等条件下直径较大的主根比细的侧根更容易发生愈伤组织;IBA和NAA激素组合更有利于不定根的诱导,其中2.00 mg/L NAA+0.05 mg/L IBA组合的诱导率达到95%。本研究结果为后续的西洋参不定根规模化培养提供重要的理论依据。In this study, fresh roots of P. quenquifolium were used as explants to characterize the induction and initiation of adventitious root. Based on MS, the formation and proliferation of adventitious root from explants were observed by supplementing with exogenous phytohormones. The results showed that the whole process of adventitious root formation in P. quinquefolium includes four stages: callus formation, callus embryogenesis,adventitious roots differentiation and development. When adventitious roots were subcultured, those with callus grew faster, and the callus grew while differentiating into new adventitious roots on their surface;In addition, the size of explants has great influence on adventitious root induction, and callus was more likely to develop from the taproots with larger diameter than the fine lateral roots under the same conditions. Combination of IBA and NAA was proved to promote the induction of adventitious roots of P. quinquefolium, and combination of 2.00 mg/L NAA+0.05 mg/L IBA could bring about the best induction rate of 95%. The results of this study would provide basis for the subsequent adventitious root culture of P. quinquefolium on a large scale.
