摘 要:目的研究罗布红麻提取物(LBHM)的急性毒性特点。方法选取16~18 g昆明小鼠,雌雄各半,急性毒性采用最大给药量法,分为对照组和LBHM最大给药量组,单次灌胃给药;亚急性毒性分为LBHM低、中、高剂量组,分别为1.2、2.4、4.8 g/(kg·d),相当于临床剂量的40、80、160倍,按照0.4 mL/10 g灌胃给药,1次/d,连续20 d。给药后进行一般观察,检测体重、脏器重量、血液生化学等指标。结果急性毒性小鼠出现活动减少、俯卧、竖毛、萎靡不振等症状,小鼠死亡率为60%;亚急性毒性给药20 d,期间中、高剂量组分别死亡1、2只小鼠;高剂量组心脏及脾脏重量、体重与对照组相比有差异(P<0.05);高剂量组肌酐明显升高(P<0.05),中剂量组尿素明显升高(P<0.05)。结论结论LBHM以最大给药量24 g/(kg·d),为临床人拟用剂量的800倍,表现出一定的急性毒性作用;亚急毒实验显示,LBHM安全剂量介于临床剂量的40~80倍之间,总体来看,LBHM使用的安全窗较高。Objective To study the acute toxicity of Apocynum extract(LBHM).Methods 16-18g Kunming mice were divided into control group and LBHM maximum dose group by maximum dosage method.Subacute toxicity was divided into LBHM low,middle and high dosage groups,which were 1.2,2.4,4.8 g/(kg·d),40,80 and 160 times of clinical dosage.The acute toxicity was administered by intragastric administration of 0.4 ml/10g once a day for 20 consecutive days.After administration,the body weight,organ weight,blood biochemistry and other indicators were measured.Results In Acute toxic mice,symptoms showed decreased activity,prone,erect hair and malaise,and the mortality rate of mice was 60%.During 20 days of sub-acute toxicity administration,1 and 2 mice died in middle and high dose groups,respectively.The weight and weight of heart and spleen in high dose group were significantly higher than those in control group(P<0.05);creatinine in high dose group was significantly higher(P<0.05),and urea in medium dose group was significantly higher(P<0.05).Conclusion The maximum dosage of LBHM is 24 g/(kg·d),which is 800 times of the intended dose for clinical use.Sub-acute toxicity test shows that the safe dosage of LBHM is between 40 and 80 times of the clinical dosage.Overall,the safe window of LBHM is high.
