摘 要:目的:观察督脉电针对阿尔茨海默病(AD)小鼠皮层细胞形态和APP、BACE1表达的影响,探讨针刺疗法抗痴呆的作用机制。方法:7月龄雄性APP/PS1小鼠共30只,随机分为模型组、电针组和药物组(药物组选用JNK特异性阻断剂SP600125),每组10只。以相同遗传背景的C57BL/6小鼠10只作为正常对照组,选取“百会”“印堂”“人中”穴,隔天针刺干预1次。治疗15次后运用苏木精-伊红(HE)染色法观察各组小鼠皮层细胞形态学的变化,采用免疫蛋白印迹法(Western blot)观察各组小鼠皮层APP、BACE1蛋白表达水平。结果:与对照组比较,模型组皮层区细胞排列紊乱,核固缩现象明显,APP、BACE1表达增多(均P<0.01);与模型组比较,电针组和药物组核固缩现象均改善,且电针组APP、BACE1表达减少(均P<0.05),药物组APP表达减少,差异无统计学意义(P>0.05),BACE1表达减少(P<0.05)。结论:督脉电针干预可能通过改善细胞核固缩现象,减少APP、BACE1的表达,降低Aβ沉积,缓解APP/PS1痴呆小鼠的学习记忆障碍。Objective:To observe the effects of Du meridian electro-acupuncture on morphology of cortical cells and the expressions of APP and BACE1 in Alzheimer's disease(AD)mice,and to explore the mechanism of anti-dementia by acupuncture.Methods:A total number of 30 male APP/PS1 mice aged 7 months were randomly divided into the model group(group M),the electro-acupuncture group(group EA)and SP600125 group(group SP),with 10 mice in each group.Ten C57BL/6 mice with the same genetic background were used as the normal control(group C).GV20,EX-HN3 and GV26 were selected for the needling.The acupuncture intervention was performed once every other day for 15 times.The morphological changes of cortical cells in each group were observed by hematoxylin-eosin(HE)staining.Western blot method was used to detect the expression levels of APP and BACE1 in the cortex of each group.Results:Compared to the group C,the cells in the cortical layer of the group M were disordered,the nuclear pyknosis was obvious,and the expressions of APP and BACE1 were increased(P<0.01).Compared to the group M,the nuclear pyknosis was improved in the group EA and in the group SP;the expressions of APP and BACE1 were decreased in the group EA(P<0.05);the expression of APP in the group SP was decreased,and the difference was not statistically significant(P>0.05),however the expression of BACE1 was decreased(P<0.05).Conclusion:Du meridian electro-acupuncture intervention may improve the learning and memory,and improve spatial exploration ability of APP/PS1 double transgenic mice by improving the nuclear pyknosis,reducing the expressions of APP and BACE1,and decreasing Aβ accumulation.
