摘 要:目的探讨葡萄籽原花青素(GSP)对糖尿病大鼠认知功能及海马细胞外信号调节激酶(ERK)/核因子E2相关性因子2(Nrf2)/血红素氧合酶1(HO-1)通路的影响。方法90只雄性大鼠随机分为正常对照组(NC)、糖尿病模型组(DM)、α-硫辛酸灌胃的阳性对照组(Con)、低剂量GSP组(L-GSP)、中剂量GSP组(M-GSP)和高剂量GSP组(H-GSP),每组各15只。采用腹腔注射STZ建立糖尿病大鼠模型。Morris水迷宫实验观察大鼠学习和记忆能力,Western blot法测定海马组织p-ERK、Nrf2、HO-1蛋白表达。结果与DM组比较,M-GSP、H-GSP组平均逃避潜伏期缩短(P<0.05),穿越平台次数增加(P<0.05),海马组织p-ERK、Nrf2、HO-1蛋白升高[(0.54±0.06)vs(0.22±0.05),(0.61±0.07)vs(0.11±0.01),(0.58±0.06)vs(0.13±0.02);(0.76±0.08)vs(0.22±0.05),(0.96±0.10)vs(0.11±0.01),(0.63±0.08)vs(0.13±0.02);P<0.05]。结论GSP可能通过激活ERK/Nrf2/HO-1通路提高糖尿病大鼠学习和记忆能力,改善大鼠认知功能。Objective To investigate the effects of grape seed procyanidin on cognitive function and hippocampal ERK/Nrf2/HO-1 pathway in chronic diabetes mellitus rats.Methods Rats were randomly divided into blank control group(NC),diabetic model group(DM),a-lipoic acid positivecontrol group(Con),low-dose grape seed procyanidin group(L-GSP),medium-dose grape seed procyanidin group(M-GSP),and high-dose grape seed procyanidin group(H-GSP).Intraperitoneal injection of streptozotocin(STZ)was used to establish the diabetic rat model.Morris water maze test was used to observe the learning and memory ability of rats.Western blot was used to detect the expressions of hippocampalp-ERK,Nrf2 and HO-1 proteins in rats.Results Compared with DM group,the average escape latency of the rats in M-GSP group and H-GSP group were shortened(P<0.05),the number of crossing platforms increased(P<0.05),the levels of p-ERK,Nrf2 and HO-1 protein in hippocampal tissue increased[(0.54±0.06)vs(0.22±0.05),(0.61±0.07)vs(0.11±0.01),(0.58±0.06)vs(0.13±0.02):(0.76±0.08)vs(0.22±0.05),(0.96±0.10)vs(0.11±0.01),(0.63±0.08)vs(0.13±0.02),P<0.05].Conclusion GSP may improve the ability of learning and memory and improve cognitive function by activating ERK/Nrf2/HO-1 pathway.
