摘 要:目的 研究白皮杉醇对急性高眼压大鼠视网膜水通道蛋白4(AQP4)表达的影响及对神经节细胞(RGCs)的保护作用。方法 将SD大鼠随机分为正常组、模型组和实验组,每组10只。用前房内生理盐水灌注法制备急性高眼压大鼠模型,正常组不予任何干预;建模后7 d,实验组灌胃给予150 mg·kg^-1白皮杉醇,正常组和模型组均灌胃给予等量生理盐水,每天1次,连续干预30 d。用眼压计测量急性高眼压大鼠建模前后眼压变化,以荧光金逆行标记及荧光显微镜观察视网膜RGCs密度,以DNA断裂的原位末端标记法(TUNEL)检测各组大鼠视网膜RGCs凋亡情况,以免疫荧光法和蛋白质印迹法检测大鼠视网膜水通道蛋白4(AQP4)表达情况。结果 药物干预结束后,正常组、模型组和实验组大鼠眼压分别为(12. 69±2. 15),(26. 98±4. 78),(20. 45±1. 96) mm Hg,大鼠视网膜RGCs密度分别为(176. 75±23. 25),(85. 44±21. 54),(123. 43±20. 84)个/视野,视网膜AQP4蛋白相对表达量分别为0. 96±0. 24,1. 52±0. 20,1. 22±0. 14,差异均有统计学意义(均P <0. 05)。结论 白皮杉醇可降低急性高眼压大鼠视网膜AQP4蛋白表达水平,抑制RGCs的凋亡,对RGCs具有较好的保护作用。Objective To study the effect of piceatannol on aquaporin 4(AQP4)expression in retina of rats with acute ocular hypertension and the protective effect of retinal ganglion cells(RGCs).Methods SD rats were randomly divided into normal group,model group and test group,with 10 rats in each group.The rats model with acute ocular hypertension were induced by perfusion of normal saline in the anterior chamber,and the normal group was given no intervention.On the 7 thday after modeling,test group was given 150 mg·kg^-1 Taxus albicans,while normal group and model group were given the same amount of normal saline once a day,for 30 d.Intraocular pressure was measured by tonometer before and after the establishment of acute ocular hypertension rat model.The retinal RGCs density was observed by fluorescence gold retrograde labeling and fluorescence microscope.The apoptosis of retinal RGCs was detected by in T-mediated UTP nick-end DNA labeling(TUNEL)and AQP4 expression were detected by immunofluorescence and Western blot.Results After drug intervention,the intra-ocular tension in normal group,model group and test group were(12.69±2.15),(26.98±4.78),(20.45±1.96)mm Hg,the RGCs of rat retina each field of vision were 176.75±23.25,85.44±21.54,123.43±20.84,the relative expression of AQP4 protein in retina were 0.96±0.24,1.52±0.20,1.22±0.14,the difference were all statistically significant(all P<0.05).Conclusion Piceatannol can decrease the expression of AQP4 protein in retina of rats with acute ocular hypertension,inhibit the apoptosis of RGCs,and has a better protective effect on RGCs.
