摘  要:目的 探究水飞蓟宾对胃癌细胞BGC-823增殖、细胞周期的调控及机制。方法 将胃癌细胞BGC-823分为空白组、阳性对照组和实验组,空白组予以等量0. 9%氯化钠,阳性对照组予以0. 1μmol·L^-1阿霉素,实验组予以200. 0μmol·L^-1水飞蓟宾。用CCK-8法检测胃癌细胞BGC-823存活率,用Annexin V-FITC/PI双染流式细胞术检测细胞周期及凋亡,用蛋白免疫印迹法检测胃癌细胞BGC-823 Cyclin E2、Cyclin B1蛋白表达量。结果 给药48 h,空白组、阳性对照组和实验组的细胞存活率分别为(100. 00±0. 00)%,(79. 09±2. 92)%,(66. 03±3. 87)%;凋亡率分别为(1. 84±0. 23)%,(36. 52±5. 49)%,(45. 56±6. 36)%;G0/G1期细胞比例分别为(62. 36±1. 23)%,(23. 96±2. 18)%,(22. 35±2. 33)%;S期细胞比例分别为(21. 53±2. 66)%,(12. 34±2. 97)%,(11. 85±2. 72)%;G2/M期细胞比例分别为(15. 28±2. 34)%,(64. 65±3. 43)%,(65. 57±3. 38)%;Cyclin E2蛋白相对表达量分别为1. 52±0. 12,0. 46±0. 15,0. 47±0. 10;Cyclin B1蛋白相对表达量分别为0. 89±0. 21,0. 29±0. 15,0. 32±0. 11,空白组分别与阳性对照组和实验组比较,差异均有统计学意义(均P <0. 05)。结论 水飞蓟宾可降低细胞周期调控蛋白Cyclin E2、Cyclin B1蛋白及基因的表达水平,将胃癌细胞阻滞于G2/M期,进而抑制细胞增殖,并诱导其凋亡。Objective To investigate the regulation and mechanism of silibinin on the proliferation and cell cycle of gastric cancer cell line BGC-823.Methods Gastric cancer cell line BGC-823 were divided into positive control group,blank group and test group.The blank group was treated with 0.9% NaCl,the positive control group was treated with0.1μmol·L^-1 adriamycin,and the test group was treated with 200.0μmol·L^-1 silibinin.CCK-8 method was used to detect the survival rate of gastric cancer cells BGC-823,Annexin V-FITC/PI double-staining flow cytometry was used to detect cell cycle and apoptosis,and Western blot was used to detect the expression of Cyclin E2,Cyclin B1 protein in gastric cancer cells.Results After 48 hours of intervention,the cell viability of blank group,positive control group and test group were(100.00±0.00)%,(79.09±2.92)%,(66.03±3.87)%,respectively;the apoptotic rate were(1.84±0.23)%,(36.52±5.49)%,(45.56±6.36)%,respectively;the percentage of G0/G1 phase cells was(62.36±1.23)%,(23.96±2.18)%,(22.35±2.33)%,respectively;the proportions of S-phase cells were(21.53±2.66)%,(12.34±2.97)%,(11.85±2.72)%,respectively;G2/M-phase cells were(15.28±2.34)%,(64.65±3.43)%,(65.57±3.38)%,respectively;the relative expression of Cyclin E2 protein was 1.52±0.12,0.46±0.15,0.47±0.10,respectively;Cyclin B1 protein was 0.89±0.21,0.29±0.15,0.32±0.11,respectively.There were statistically significant differences between the blank group and the positive control/experimental group(all P<0.05).Conclusion Silibinin can reduce the expression levels of cyclin-regulating protein Cyclin E2,Cyclin B1 protein and m RNA,block gastric cancer cells at G2/M phase,and then inhibit cell proliferation and induce apoptosis.
