摘  要:目的探讨线粒体功能相关基因在类风湿关节炎(RA)滑膜组织中的表达变化及其与滑膜组织病理变化的关联。方法利用来自GEO数据库的基因表达芯片数据,通过生物信息学方法分析关节滑膜组织中线粒体功能相关基因的表达情况,并对这些基因进行功能、通路注释与相互作用网络分析。结果与正常对照相比,RA患者存在1 986个显著差异表达的基因,其中169个基因的编码蛋白定位于线粒体并与线粒体结构、功能相关。功能注释分析结果显示:异常表达的线粒体功能相关基因的功能富集通路主要集中于线粒体的核苷酸代谢(GO:0009117)、线粒体结构(GO:0007005)等功能通路,并激活了部分炎症反应通路(GO:2001233、GO:0034976、GO:0006979和GO:0072593等)。而其蛋白-蛋白相互作用网络中的基因则富集了线粒体相关蛋白的加工(HSA04141)、核苷酸代谢(GO:0009117、GO:0006753)、翻译(R-HSA-1799339、R-HSA-72766)、修饰(R-HSA-446203、HSA00510)、小分子转运(R-HSA-382551)和体液水平的调控(GO:0050878)功能通路。结论差异表达的线粒体功能相关基因与关节滑膜组织的线粒体功能异常及RA的病理变化密切相关。Objective To investigate the association between expression changes of mitochondrial-related genes in synovial tissues of rheumatoid arthritis( RA) patients and pathological changes in synovial tissues. Methods Based on public microarray-based transcriptome data from GEO database,bioinformatics methods were used to analyze the expression of the genes associated with mitochondrial function in synovial tissues and to analyze the function,pathway annotation and interaction network of these genes. Results A total of 1 986 genes were identified with significant differences when compared with normal controls,among which,169 genes encode mitochondrial proteins and are associated with the structure and function of mitochondria. Functional annotation analysis showed that the functional enrichment pathways of aberrantly expressed mitochondrial function-related genes were mainly focused on functional pathways such as nucleotide metabolic process( GO: 0009117) and mitochondrion structure( GO: 0007005),and some activated inflammatory reaction pathways( GO: 2001233,GO: 0034976,GO: 0006979 and GO: 0072593,etc.). The genes in the protein-protein interaction network were enriched in the processing of mitochondrion-associated proteins( HSA04141),nucleotide metabolism( GO: 0009117,GO: 0006753),translation( R-HSA-1799339,R-HSA-72766),protein modification( R-HSA-446203,HSA00510),small molecule transport( R-HSA-382551) and regulation of humoral levels( GO: 0050878) functional pathway. Conclusion Differentially expressed mitochondria-related genes are closely associated with the abnormal mitochondrial function in synovial tissue and pathology of RA.
