摘  要:【目的】探讨单磷酰脂质A(monophosphoryl lipid A,MPLA)对口蹄疫(food and mouth disease,FMD)O型灭活疫苗的免疫增强效应,提高口蹄疫灭活疫苗的免疫效果.【方法】通过检测MPLA刺激脾脏细胞的增殖情况和刺激DC后成熟及吞噬能力的大小,对MPLA的体外免疫刺激效应进行评价;将MPLA与ISA 206和灭活FMDVO型抗原配制疫苗后免疫小鼠,通过检测抗体水平、T细胞亚群、抗原特异性淋巴细胞增殖及浆细胞的比例,评价MPLA对口蹄疫O型灭活疫苗的免疫增强作用.【结果】MPLA体外可刺激B淋巴细胞增殖,上调DC表面共刺激分子的表达,使其吞噬能力减弱;制备疫苗免疫小鼠,血清抗体水平检测发现,MPLA组在7、14 d时抗体水平显著高于ISA 206组,且其抗原特异性的B淋巴细胞增殖,外周血中CD19+CD27+CD38+浆细胞的比例也显著高于ISA 206组.【结论】MPLA通过促进B淋巴细胞的增殖与活化增加了抗体的产生,增强了小鼠对口蹄疫疫苗的免疫应答能力,证实MPLA是口蹄疫疫苗的良好候选免疫佐剂.【Objective】The immune enhancement effect of monophosphoryl lipid A(MPLA)on serotype O food and mouth disease(FMD)inactivated vaccine was studied in order to enhance the immune effect of FMD inactivated vaccine.【Method】The effect of MPLA on immune stimulation in vitro was evaluated by detecting the proliferation of spleen cells,and postmaturation and phagocytosis of stimulated DC cells.Mice were immunized with the vaccine prepared by MPLA,ISA 206 and inactivated O type FMDV antigen,and then the immune enhancement effect was evaluated by detecting the antibody level,T cell subgroup,antigen-specific lymphocyte and the ratio of plasma cells in peripheral blood.【Result】MPLA stimulated the proliferation of B lymphocytes and up-regulated the expression of co-stimulatory molecules on DC surface to weaken the phagocytic ability in vitro.The vaccine-immunized mice were tested for serum antibody levels,and the antibody levels in the MPLA group were significantly higher than 206 group on 7 th and 14 th day;The ratio of antigen-specific B lymphocyte proliferation and CD19+CD27+CD38+plasma cells in peripheral blood were also significantly higher than 206 group.【Conclusion】MPLA could enhance the production of antibody by promoting the proliferation and activation of B lymphocytes to strengthen the immune response of mice for foot-and-mouth disease vaccine.Therefore,MPLA could be used as a promising candidate immune adjuvant for foot-and-mouth disease vaccine.
