摘 要:目的:探究连翘苷(Phillyrin)对糖尿病肾病大鼠肾功能、炎症反应和氧化应激反应的影响以及作用机制。方法:高糖高脂加链脲菌素复制糖尿病肾病大鼠模型。将糖尿病肾病大鼠随机分为:糖尿病肾病组,连翘苷5 mg/kg组,连翘苷10 mg/kg组,连翘苷20 mg/kg组,另外设置正常对照组。苏木伊红染色检测肾组织病理损伤。全自动生化分析仪检测肾功能指标。酶联免疫检测肾组织中炎症因子的含量;免疫组化检测肾组织中转化生长因子β1(TGF-β1)、Ⅳ胶原(Col-Ⅳ)、纤连蛋白(Fibronectin)的表达水平;检测肾组织中氧化应激相关指标的含量。结果:糖尿病肾病大鼠经连翘苷治疗后,肾组织中肾小球肿胀、基底膜增厚明显减轻;尿素氮、肌酐、尿酸和尿白蛋白的含量显著减少;肾组织中白细胞介素4(IL-4)、IL-6、IL-18、IL-1β、肿瘤坏死因子α(TNF-α)、单核细胞趋化蛋白1(MCP-1)的含量显著升高;肾组织中TGF-β1、Col-IV、Fibronectin的表达水平显著降低;肾组织中总超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)的活性均显著升高,丙二醛(MDA)、氧化性谷胱甘肽(GSSG)的含量显著降低,GSH的含量显著升高;且上述作用具有连翘苷剂量依赖效应。结论:连翘苷抑制糖尿病肾病大鼠的炎症反应,缓解肾组织损伤,对糖尿病肾病大鼠具有保护作用,且作用机制与抑制TGF-β1的表达相关。Objective:To investigate the effects of phillyrin on renal function,inflammation and oxidative stress in diabetic nephropathy rats.Methods: Diabetic nephropathy rats were induced by high glucose,high fat and streptozotocin.Diabetic nephropathy rats were randomly divided into diabetic nephropathy group,phillyrin 5 mg/kg group,phillyrin 10 mg/kg group,phillyrin 20 mg/kg group and normal control group.Hematoxylin eosin staining was used to detect pathological damage of kidney tissue.Automatic biochemical analyzer was used to detect renal function.Enzyme-linked immunosorbent assay(ELISA) was used to detect the levels of inflammatory factors in kidney tissues,immunohistochemistry was used to detect the expression of TGF-β1,Col-Ⅳ and Fibronectin,and the content of oxidative stress-related indicators in kidney tissues.Results: After treatment with forsythiasin,the glomerular swelling and basement membrane thickening were significantly reduced,the contents of urea nitrogen,creatinine,uric acid and urinary albu min were significantly decreased,the contents of IL-4,IL-6,IL-18,IL-1β,TNF-α and MCP-1 were significantly increased,and the contents of TGF-β1,Col-Ⅳ,Fibronectin in renal tissue were significantly increased.The activity of SOD,CAT and GPx in kidney tissues were significantly increased,the content of MDA and GSSG were significantly decreased,and the content of GSH was significantly increased.Conclusion: Phillyrin can inhibit inflammation and alleviate renal injury in diabetic nephropathy rats,and has protective effect on diabetic nephropathy rats.The mechanism is related to the inhibition of TGF-β1 expression.
