摘 要:目的观察越鞠丸对功能性消化不良(FD)模型大鼠胃肠动力、血清胃动素(MTL)水平及胃窦黏膜一氧化氮(NO)含量的影响。方法取48只健康雄性SD大鼠,随机分为空白组、模型组、莫沙必利组、越鞠丸低、中、高剂量(4.5g/kg、9g/kg、18g/kg)组,每组8只。各组大鼠分别予以对应药物连续治疗7天后,观察大鼠体质量、饮水量、进食量、胃内残留率与小肠推进率的改变;检测胃窦黏膜NO表达和血清MTL含量。结果与模型组比较,越鞠丸各剂量组均出现进食量、饮水量增加,莫沙必利组、越鞠丸中、高剂量组大鼠胃内残留率减少[(63.89±16.74)%、(56.79±20.52)%、(64.25±17.79)%比(81.18±6.78)%,P均<0.05)];莫沙必利组、越鞠丸各组大鼠小肠推进率升高[(59.47±3.76)%、(52.28±2.75)%、(56.06±1.31)%、(59.74±3.02)%比(46.99±4.59)%,P均<0.05)]。与模型组比较,莫沙必利组和越鞠丸中、高剂量组大鼠血清MTL升高[(202.64±27.64)pg/mL、(181.78±35.15)pg/mL、(214.92±40.01)pg/mL比(124.35±10.43)pg/mL,P均<0.05];胃窦黏膜组织NO含量减少[(0.09±0.06)μmol/L、(0.11±0.01)μmol/L、(0.09±0.08)μmol/L比(0.17±0.03)μmol/L,P均<0.05]。结论越鞠丸能加快大鼠胃肠动力,其作用机制可能与上调MTL水平、下调NO水平有关。Objective To observe the effects of Yueju pill on gastrointestinal motility, serum motilin(MTL) level and nitric oxide(NO) content in antral mucosa of rats with functional dyspepsia(FD). Methods Forty-eight male SD rats were randomly divided into blank group, model group, mosapride group, groups of the low(4.5 g/kg), medium(9 g/kg) and high(18 g/kg) dose of Yueju pill, with 8 animals in each group. After 7 days of continuous treatment with corresponding drugs in each group, the changes of body weight, water intake, food intake, gastric residual rate and small intestinal propulsion rate were observed, and the expression of NO in antral mucosa and the content of MTL in serum were detected. Results Compared to the model group, rats in different Yueju pill groups showed an increase in food intake and water intake. The gastric residual rate of rats in the mosapride group, the medium and high dose group decreased, i.e.,(63.89±16.74)%,(56.79±20.52)% and(64.25±17.79)% vs(81.18±6.78)%(P<0.05, respectively);the small intestinal propulsion rate of rats in the mosapride group and all Yueju pill groups increased, i.e.,(59.47±3.76)%,(52.28±2.75)%,(56.06±1.31)% and(59.74±3.02)% vs(46.99±4.59)%(P<0.05, respectively);the serum MTL of rats in the mosapride group, the medium and high dose groups increased, i.e.,(202.64±27.64),(181.78±35.15) and(214.92±40.01) vs(124.35±10.43)pg/mL(P<0.05, respectively), and the content of NO in antral mucosa decreased, i.e.,(0.09±0.06),(0.11±0.01) and(0.09±0.08) vs(0.17 ±0.03)mol/L(P<0.05, respectively). Conclusion Yueju pill can accelerate gastrointestinal motility in rats. Its mechanism may be related to up-regulation of MTL level and down-regulation of NO level.
