森林草莓‘Hawaii 4’继代培养与遗传转化条件的优化

摘  要:【目的】探明森林草莓高效的组织培养和基因工程转化方法。【方法】以森林草莓组培苗为试验材料,对继代培养条件包括基本培养基、培养基的pH值、含糖量、琼脂量等以及遗传转化条件包括植物激素的选择、农杆菌的浓度等进行了摸索。【结果】组培苗继代培养的最佳条件为:1/2 MS,10g/L糖,7g/L琼脂粉,pH5.8。遗传转化的最佳条件为:叶片叶龄:39d左右,菌液浓度OD600=0.6,侵染时长15min,植物激素种类与配比:3.0mg/L的6-BA和0.2mg/L的IBA,暗培养2d,草铵膦筛选质量浓度:2mg/L。【结论】获得了有利于森林草莓愈伤组织生芽和幼苗生长的优化培养基成分,以及高效率的基因转化条件。【Objective】To explore the appropriate conditions for subculture and genetic transformation for woodland strawberry’Hawaii 4’.【Methods】The conditions for subculture of woodland strawberry such as the concentration of basic MS medium,pH,sugar content,agar amount,etc.,and for the genetic transformation including hormones,the concentration of Agrobacterium tumefaciens,etc.were systematically explored.【Results】The appropriate conditions for subculture were:1/2 MS,10 g/L sugar,7 g/L agar,pH 5.8.The optimal conditions for the transformation were as follows:39-day-old leaves were cultured in medium containing 3.0 mg/L 6-BA and 0.2 mg/L IBA for 2 days;and the optimal screening concentration was 2 mg/L herbicides.【Conclusion】The appropriate subculture conditions and genetic transformation conditions for the woodland strawberry’Hawaii 4’were established.
