摘  要:目的研究基质金属蛋白酶的抑制因子TIMP-1蛋白糖基化及糖链异常在子宫内膜中的特征性改变,探讨TIMP-1在子宫内膜异位症的发病和发展过程中的意义。方法使用蛋白质印迹法检测43例子宫内膜异位症的异位内膜和在位内膜以及20例健康子宫的内膜中TIMP-1蛋白的糖基化及糖链异常水平。结果在EMs异位内膜中TIMP-1蛋白高度糖基化,相较于子宫内膜异位症的在位内膜和健康子宫内膜,统计学意义上有显著性差异(P<0.05)。异位的内膜组织里TIMP-1蛋白的表达不呈现周期的改变,但是同r AFS临床分期有关联(P<0.05)。结论异位内膜组织中TIMP-1高度糖基化,导致MMP-1/TIMP-1平衡失调,增强了异位内膜细胞的侵袭能力,在子宫内膜异位症的发病和发展过程中发挥重要影响。Objective To study the glycosylation of matrix metalloproteinase inhibitor TIMP-1 protein and the characteristic changes of abnormal sugar chain in endometrium,and to discuss the significance of TIMP-1 in the pathogenesis and development of endometriosis.Methods The glycosylation and abnormal sugar chain levels of TIMP-1 protein in 43 cases of ectopic and eutopic endometrium of endometriosis and 20 cases of healthy endometrium were detected by Western blotting.Results TIMP-1 protein was highly glycosylated in ectopic endometrium of EMs,which was significantly different from eutopic endometrium and healthy endometrium(P<0.05).The expression of TIMP-1 protein in ectopic endometrium did not change periodically,but was associated with the clinical stage of R AFS(P<0.05).Conclusion The high glycosylation of TIMP-1 in ectopic endometrium leads to imbalance of MMP-1/TIMP-1,enhances the invasive ability of ectopic endometrial cells,and plays an important role in the pathogenesis and development of endometriosis.
