摘  要:目的探讨巴豆醛亚慢性染毒致雄性大鼠肺损伤作用,分析其损伤的可能机制。方法将40只SPF级雄性Wistar大鼠随机分为对照组和2.5、4.5、8.5 mg/kg 3个染毒组,每组各10只,染毒组分别给予2.5、4.5、8.5 mg/kg巴豆醛溶液连续灌胃染毒120 d,每天1次,对照组给予等量生理盐水。染毒结束后测定大鼠体重,颈椎脱臼处死后迅速分离肺组织,计算脏器系数并进行组织病理学检查;ELISA法测定肺组织中白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)水平;测定血清中丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)含量。结果与对照组比较,4.5、8.5 mg/kg染毒组大鼠体重增加较慢,肺脏重量及脏器系数均下降,差异均有统计学意义(P<0.05)。染毒组大鼠肺组织结构紊乱,肺泡壁增厚,可见炎性细胞浸润。与对照组比较,4.5、8.5 mg/kg染毒组大鼠肺组织中IL-6、IL-1β水平明显上升,2.5、4.5、8.5 mg/kg染毒组大鼠肺组织中TNF-α水平明显上升,差异均有统计学意义(P<0.05);与对照组比较,4.5、8.5 mg/kg染毒组大鼠血清中MDA活力升高,SOD、GSH-PX活力降低,差异均有统计学意义(P<0.05)。结论巴豆醛可能通过上调肺组织中炎性因子表达,改变氧化平衡状态,促使大鼠发生炎性及氧化应激损伤。Objective To observe the lung injury of male rats induced by sub-chronic exposure to crotonaldehyde,and to explore the possible mechanism of injury.Methods Forty SPF male Wistar rats were randomly divided into control group and 3 groups in each group,and each group received 0.0,2.5,4.5,8.5 mg/kg body weight crotonaldehyde solution for continuous intragastric administration.120 d,once a day.After the end of the exposure,the body weight of the rats was measured,and the lung tissues were quickly separated after cervical dislocation.The organ coefficients were calculated and histopathological examination was performed to determine malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione.Peroxidase(GSH-Px)content;ELISA was used to measure interleukin(IL)-6,IL-1β,and tumor necrosis factor(TNF)-αin lung tissues.Results Compared with the control group,the weight gain of the rats in the 4.5 and 8.5 mg/kg exposure groups was small,and the lung weight and organ coefficient of the exposed group decreased,the difference was statistically significant(P<0.05).In the exposed group,the lung tissue structure was disordered,the alveolar wall was thickened,and inflammatory cell infiltration was observed.Compared with the control group,the MDA activity in the serum of the rats in the 4.5 mg/kg and 8.5 mg/kg groups increased,and the SOD and GSH-Px activities decreased,the difference was statistically significant(P<0.05).TNF-αlevels in the lung tissues of rats exposed to 4.5 mg/kg and 8.5 mg/kg,and levels of(IL)-6 and IL-1βin the lungs of rats in the 2.5,4.5,and 8.5 mg/kg groups.Significantly increased,the difference was statistically significant(P<0.05).Conclusion Crotonaldehyde may induce inflammatory and oxidative stress damage in rats by up-regulating the expression of inflammatory factors in lung tissue and changing the oxidative balance.
