摘 要:目的:观察三七总皂苷(PNS)对脑缺血再灌注损伤小鼠海马Bax和Bcl-2表达的影响。方法:采用双侧颈总动脉夹闭法构建缺血再灌注损伤模型,选用健康的6周龄昆明小鼠33只,分为空白组、PNS组、缺血9d组(缺血再灌注9d后取脑)、缺血24h组(缺血再灌注24h后取脑)。PNS组于缺血前3d开始腹腔注射PNS[30mg/(kg·d)],直至再灌注9d后取脑,缺血9d组和缺血24h组予以等体积0.9%氯化钠注射液腹腔注射。各组取脑后制作冰冻切片,采用免疫组织化学法检测小鼠海马区脑组织Bax和Bcl-2阳性表达细胞数。结果:与空白组比较,缺血9d组Bcl-2、Bax水平均明显增高,差异有统计学意义(P<0.05)。与缺血9d组比较,PNS组Bax的表达水平平均值明显降低,Bcl-2/Bax的表达水平明显增高,差异均具有统计学意义(P<0.05);缺血9d组Bax和Bcl-2的表达均较缺血24h组增高,差异具有统计学意义(P<0.05)。结论:PNS对短暂性双侧颈动脉闭塞型小鼠缺血再灌注损伤的作用机制是早期通过降低Bax表达,增大Bcl-2/Bax比值,抑制神经元凋亡,从而对大脑海马神经元起保护作用,且再灌注第9天损伤程度可能更大。Objective:To investigate the effect of Panax notoginseng saponins(PNS)on the expression of Bax and Bcl-2 in the hippocampus in mice with cerebral ischemia/reperfusion injury.Methods:A mouse model of cerebral ischemia/reperfusion injury was established by clipping of both common carotid arteries.A total of 33 healthy Kunming mice aged 6 weeks were divided into blank group,PNS group,9-day ischemia group(brain tissue was collected after 9 days of ischemia/reperfusion),and 24-hour ischemia group(brain tissue was collected after 24 hours of ischemia/reperfusion).The mice in the PNS group were given intraperitoneally injected PNS30 mg/kg·d since day 3 before ischemia,and brain tissue was collected after 9 days of reperfusion;those in the 9-day ischemia group and the 24-hour ischemia group were given intraperitoneal injection of an equal volume of0.9%sodium chloride injection.After brain tissue was collected for the preparation of frozen sections,immunohistochemistry was used to measure the number of cells with the expression of Bax and Bcl-2 in the hippocampus.Results:Compared with the blank group,the 9-day ischemia group had a significant reduction in the mean level of Bcl-2 and Bax(P<0.05).Compared with the 9-day ischemia group,the PNS group had a significant reduction in the mean level of Bax and a significant increase in Bcl-2/Bax ratio(P<0.05).The 9-day ischemia group had significantly higher expression of Bax and Bcl-2 than the 24-hour ischemia group(P<0.05).Conclusion:In the mouse model of transient ischemia-reperfusion injury due to occlusion of both carotid arteries,PNS exerts a protective effect on hippocampal neurons by reducing Bax expression,increasing Bcl-2/Bax ratio,and inhibiting neuron apoptosis in the early stage,and there may be a relatively high degree of injury on day 9 of reperfusion.
