摘  要:建立高效液相色谱法测定3-甲氧基-4-羟基苄胺盐酸盐(香草胺盐酸盐)的方法。色谱柱为C8柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.25%高氯酸水溶液(20∶80),流量为1.0 mL/min,检测波长为230 nm,柱温为25℃。香草胺盐酸盐的质量浓度在5.25~262.5μg/mL范围内与色谱峰面积线性关系良好(r2=1.0),测定结果的相对标准偏差为0.09%(n=6),检出限为16.8 ng/mL,定量限为84 ng/mL,加标回收率为97.78%~100.71%。该方法简便,准确,专属性强,可用于香草胺合成过程的质量控制。A method for the determination of 3-methoxy-4-hydroxybenzylamine hydrochloride(vanilline hydrochloride)was established by high performance liquid chromatography(HPLC).The chromatographic column was C8(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.25%perchloric acid aqueous solution(20∶80),the flow rate was 1.0 mL/min,the detection wavelength was 230 nm,and the column temperature was 25℃.In the range of 5.25-262.5μg/ml,vanilline hydrochloride has a good linear relationship with the peak area(r2=1.0).The relative standard deviation of the determination results was 0.09%(n=6),the detection limit was 16.8 ng/mL,the quantitative limit was 84 ng/mL.and the recovery was 97.78%-100.71%.The method is simple,accurate and specific,and it can be used for the quality control of vanillin synthesis.
