摘 要:为了研究不同月龄对幼龄吕梁黑山羊耳缘成纤维细胞增殖速度的影响,探索最佳的供体年龄,笔者采用组织块培养法建立细胞系,观察细胞形态,测定细胞首次传代时间、平均传代时间及复苏后平均传代时间,分析不同月龄对吕梁黑山羊耳缘成纤维细胞体外培养的影响。试验结果表明,年龄对细胞形态无影响,但幼龄吕梁黑山羊的月龄会影响首次观察到细胞从组织块迁出的时间、细胞首次传代时间、平均传代时间及复苏后平均传代时间(P<0.05);冻存对3月龄供体的细胞增值速度无明显影响(P>0.10)。说明选择3月龄的吕梁黑山羊个体可以通过组织块法建成稳定的成纤维细胞系。本试验使吕梁黑山羊种质资源在细胞水平得到保存,为吕梁黑山羊成纤维细胞系的建立以及开展其种质资源的保存提供资料。In order to study the effect of the age of young Lvliang Black Goat on the proliferation rate of fibroblasts in the ear, and explore the optimal donor age, test using tissue culture method to establish a cell line, observation of cell morphology, determination of extend the time for the first time, extend the average average extend time after time and recovery, analysis of the edge of the ear for Lvliang Black Goat fibroblasts in vitro.The results showed that age had no effect on cell morphology.However, the monthly age of the young Lvliang Black Goat would affect the time of the first observed cell migration from the tissue block, the time of the first cell passage, the average passage time and the average passage time after resuscitation(P<0.05);cryopreservation had no significant effect on cell proliferation rate of 2-and 3-month-old donors(P>0.10). It indicates that the selected individuals of Lvliang Black Goat aged 2 or3 months can build stable fibroblast cell lines by tissue block method.This experiment allowed the germplasm resources of Lvliang Black Goat to be preserved at the cellular level, which provided a reference for the establishment of Lvliang Black Goat fibroblast cell line and the preservation of its germplasm resources.
