摘  要:目的探究周期性射频辐射(RF)对果蝇睡眠行为的影响及调控机制。方法实时连续监测果蝇在光照、RF(1800 MHz,1 W/m2,12 h/d)及黑暗条件下的行为变化,对比分析各条件下果蝇运动和睡眠的异同。收集处理2 h后的样品,分析RF对果蝇生物钟基因per、tim、clk、cyc和cry及神经递质关键合酶编码基因hdc、gdh、gad和trh表达水平的影响。结果光照组和RF组的运动和睡眠均表现出与处理周期一致的节律变化,黑暗组节律振幅减弱;与黑暗组相比,光照组果蝇白天和夜晚的睡眠质量均显著上升,RF组果蝇白天和黑暗组的表现一致,但夜间睡眠时间显著延长,睡眠质量上升。短时间暴露下,光照组和RF组果蝇上述5个生物钟基因表达水平全部下调,只是光照组clk、RF组tim下调更显著。此外,光照组和RF组hdc和gad基因表达水平上调,gdh下调,RF组trh基因下调。结论与光同步的周期性RF可能通过影响生物钟和神经递质相关合成基因的表达,进而影响果蝇的睡眠节律、时间及质量,在昼夜节律调控方面可能具有类似光照的功能。Objective To investigate the effect of periodic radiofrequency(RF)radiation on sleep behavior of Drosophi la melanogaster and the regulatory mechanism.Methods The behavioral changes of D.melanogaster under light,RF radi ation(1800 MHz,1 W/m^2,12 h/d)and dark conditions were continuously monitored in real time.The similarities and dif ferences between D.melanogaster movement and sleep under different conditions were compared and analyzed.Samples were collected for 2 h to analyze the effects of RF radiation on the expression levels of clock genes,such as per,tim,clk,cyc and cry,and the neurotransmitter key synthase encoding genes hdc,gdh,gad and trh.Results The movement and sleep of the illumination group and the RF group showed rhythm changes consistent with the treatment cycle,and the rhythm amplitude in the dark group was weakened.Compared with the dark group,the sleep quality of the light group in creased significantly during the day and night.As for the RF group,the performance of D.melanogaster during the day was consistent with that of the dark group,but the nighttime sleep time was significantly prolonged and the quality of sleep im proved.Under the short term exposure,the expression levels of the above five clock genes in the light and RF groups were all down regulated except that the clk and tim were down regulated to different extents.In addition,the expression levels of hdc and gad genes were up regulated in the light and RF groups,gdh genes were up regulated,and the expression levels of trh gene was down regulated in the RF group.Conclusion Periodic RF radiation synchronized with light may affect the expression of circadian clock and neurotransmitter related synthetic genes,hereby affecting the rhythm,time and quality of sleep of the D.melanogaster,and may perform a similar illumination function in the regulation of circadian rhythm.
