乌头注射液对膝骨性关节炎兔关节液TNF-α、PGE2含量的影响

摘　　要：目的:观察乌头注射液膝关节腔注射对膝骨性关节炎兔关节液中TNF-α(肿瘤坏死因子)和PGE 2(前列腺素E2)含量的影响,探讨乌头注射液对膝骨性关节炎作用机制。方法:将40只新西兰大耳白兔随机分为模型组、乌头注射液组、玻璃酸钠组和正常对照组。在除正常组以外的其他3组兔膝关节腔中注射木瓜蛋白酶建立膝骨性关节炎模型。实验造模完成后,分别以0.1 mL/kg的剂量标准依次注入玻璃酸钠、乌头注射液和生理盐水,药品注射时间为第1、4、7、10天。注射完毕,饲养1周后以空气栓塞法处死所有实验白兔。抽取各组兔膝关节液,采用ELISA法测定关节液TNF-α和PGE 2的含量。结果:与正常组比较,模型组兔关节液TNF-α和PGE 2含量明显升高(P<0.05);与模型组比较,乌头注射液组和玻璃酸钠组两者均明显下降(P<0.05),但高于正常组(P<0.05);乌头注射液组两者均较玻璃酸钠组降低,组间差异具有统计学意义(P<0.05)。结论:乌头注射液对膝骨性关节炎的治疗作用机制可能与抑制TNF-α和PGE 2的表达有关。Objective:To investigate the mechanism of Wutou Injection in the treatment of knee osteoarthritis by injecting it into the knee joint cavity of rabbits with knee osteoarthritis and observing the changes of TNF-αand PGE 2 contents in articular fluid.Methods:40 New Zealand white rabbits were randomly divided into model group,Wutou Injection group,Sodium hyaluronate group and normal control group.In addition to the normal group,rabbits of the other three groups were injected papain in knee cavity to establish the model of knee osteoarthritis.After the experimental modeling was completed,the rabbits in the sodium hyaluronate group,Wutou Injection group and model group were respectively injected with sodium hyaluronate,Wutou Injection and saline at the standard dose of 0.1 mL/kg on the first,fourth and seventh day.After injection,all rabbits were sacrificed by air embolization after 1 week.The contents of TNF-αand PGE 2 in rabbits’articular fluid were determined by ELISA method.Results:The contents of TNF-αand PGE 2 in articular fluid of model group rabbits were increased,they were higher than those in normal group(P<0.05),the contents of TNF-αand PGE 2 in articular fluid of sodium hyaluronate group and Wutou Injection group were decreased significantly,they were lower than those of model group(P<0.05),there were statistical significance between the sodium hyaluronate group and Wutou Injection group(P<0.05).Conclusion:The mechanism of Wutou Injection on knee osteoarthritis is related to the inhibition of TNF-αand PGE 2 expressions in knee joint fluid.
