摘 要:目的探讨系统性红斑狼疮(SLE)全血样本线粒体功能相关基因的差异表达,旨在为探索SLE的发病机制提供新思路。方法从NCBI的GEO数据库中下载SLE全血转录组测序数据(GSE72509),借助R软件,利用t检验和差异倍数法筛选差异表达基因,并对与SLE线粒体功能相关的差异表达基因进行功能注释、功能富集和蛋白质互作网络分析。结果本研究共从SLE全血样本中筛选出1478个差异表达基因。其中,SLE差异表达的线粒体相关基因有40个,主要与氨基酸及其衍生物的代谢有关;SLE差异表达的线粒体基因有8个,主要与氧化磷酸化和ATP代谢过程有关。结论线粒体功能相关基因的表达失调与SLE的发生、发展有关。Objective To investigate the differential expression of mitochondrial function-related genes in whole blood samples from patients with systemic lupus erythematosus(SLE),and to provide new ideas for studying the pathogenesis of SLE.Methods Transcriptome sequencing data of whole blood samples from SLE patients and healthy controls(GSE72509)were downloaded from NCBI GEO database.Differentially expressed genes(DEGs)were screened by using the methods of t-test and FC(Fold Change)with R software.Besides,functional annotation,functional enrichment and protein interaction network analysis were performed with mitochondria-related DEGs.Results A total of 1478 DEGs were identified.Among them,40 DEGs were mitochondria-related genes of SLE and they were mainly involved in the metabolism of amino acids and their derivatives.In addition,8 differentially expressed mitochondrial genes were identified to be associated with SLE and they were mainly involved in oxidative phosphorylation and ATP metabolism.Conclusion The expression dysregulation of mitochondrial function may be associated with the occurrence and development of SLE.
