摘 要:目的观察不同浓度五味子乙素对高糖诱导人脐静脉内皮细胞(HUVEC)的抗衰老作用,并探讨其机制。方法将HUVEC分为6个组。正常组给予5.5 mmol/L的D-葡萄糖,其余各组均给予25 mmol/L的D-葡萄糖;五味子乙素1、2、3、4组在高糖诱导基础上,再分别给予5、10、15、20μmol/L的五味子乙素。各组细胞传代20代,采用衰老相关的β-半乳糖苷酶(SAβ-gal)染色法评估细胞衰老情况。采用流式细胞仪分析各组细胞周期,检测各组细胞中活性氧簇(ROS)、一氧化氮(NO)水平。结果高糖组、五味子乙素1组、五味子乙素2组、五味子乙素3组衰老细胞百分比高于正常组;五味子乙素各组衰老细胞百分比低于高糖组,五味子乙素不同浓度组间差异均有统计学意义(P均<0.01)。高糖组、五味子乙素1组、五味子乙素2组、五味子乙素3组G0/G1期细胞比例高于正常组,S期细胞比例低于正常组;五味子乙素各组G0/G1期细胞比例低于高糖组,S期细胞比例高于高糖组;五味子乙素不同浓度组间差异均有统计学意义(P均<0.01)。高糖组、五味子乙素1组、五味子乙素2组、五味子乙素3组ROS水平高于正常组;五味子乙素各组ROS水平低于高糖组;五味子乙素不同浓度组间差异均有统计学意义(P均<0.01)。高糖组、五味子乙素1组、五味子乙素2组、五味子乙素3组NO水平低于正常组;五味子乙素各组ROS水平高于高糖组;五味子乙素不同浓度组间差异均有统计学意义(P均<0.01)。结论五味子乙素可延缓高糖诱导的HUVEC衰老进程,该作用可能与其减轻氧化应激损伤有关。Objective To observe the anti-aging effects of different concentrations of Schisandrin B on high glucose-induced human umbilical vein endothelial cells(HUVECs)and to explore its mechanism.Methods The HUVECs were divided into 5 groups.The HUVECs in the normal group were given 5.5 mmol/L D-glucose,and the other groups with 25 mmol/L D-glucose;the HUVECs in the Schisandrin B 1,2,3,and 4 groups were given 5,10,15 and 20μmol/L Schisandrin B,respectively,on the basis of high glucose induction.HUVECs in the each group were passaged for 20 generations.Cell senescence was assessed using senescence-associated β-galactosidase(SAβ-gal)staining.The cell cycle of each group was analyzed by flow cytometry.The levels of reactive oxygen species(ROS)and nitric oxied(NO)in each group were examined.Results The percentage of senescent cells in the high glucose group,the Schisandrin B 1,2,and 3 groups was higher than that in the normal group;the percentage of senescence cells in the Schisandrin B groups was lower than that in the high glucose group,with statistically significant difference between different concentrations of Schisandrin B groups(all P<0.01).The proportion of cells in the G 0/G 1 phase of the high glucose group,Schisandrin B 1,2,and 3 groups was higher than that in the normal group,and the proportion of cells in S phase was lower than that in normal group;the proportion of cells in the G 0/G 1 phase of Schisandrin B groups was lower than that of high glucose group,the proportion of cells in S phase was higher than that in high glucose group,with statistically significant difference between different concentrations of Schisandrin B groups(all P<0.01).The levels of ROS in the high glucose group,the Schisandrin B 1,2,and 3 groups were higher than those in the normal group;the ROS levels in the Schisandra B groups were lower than those in the high glucose group,with statistically significant difference between different concentrations of Schisandrin B groups;the levels of NO in the high glucose group,the Schisandra B 1,
