摘 要:目的建立HPLC法测定绿豆中牡荆素的含量。方法采用Symmetry■ C18色谱柱(150 mm×4.6 mm,5μm),以甲醇-0.3%磷酸水(35∶65)为流动相等度洗脱,流速:1 ml/min,检测波长:260 nm,柱温:25℃。结果牡荆素进样量在0.04~0.44μg(r=0.9999)范围内与峰面积呈良好的线性关系,平均回收率为99.34%,RSD为1.37%(n=6),不同产地绿豆中牡荆素含量范围为1.04~1.71 mg/g。结论该方法准确、可靠、重复性好,可用于绿豆的质量控制。Objective To establish an HPLC method for the determination of vitexin in seeds of Vigna radiata.Methods The analysis was performed on a Symmetry C18 column(150 mm×4.6 mm,5μm)at 25℃,using methanol-0.3%phosphoric acid(35∶65)as the mobile phase at a flow rate of 1.0 ml/min with the UV detection wavelength at 260 nm.Results The linear range of vitexin was 0.04~0.44μg(r=0.9999).The average recovery was 99.34%with RSD of 1.37%(n=6).The content of vitexin in seeds of Vigna radiate from different was 1.04~1.71 mg/g.Conclusion The established method is accurate,reliable and reproducible in the determination of vitexin in seeds of Vigna radiata,and can be used for the quality control of seeds of Vigna radiata.
