摘 要:为了研究异氟醚对7日龄幼鼠神经认知功能的影响及推测损伤的机制,将45只7日龄昆明系小鼠,随机分为A(1.5%异氟醚1 h)、B(1.5%异氟醚1.5 h)、C(1.5%异氟醚2 h)、D(2%异氟醚2 h)及Control(对照组)5组。使用异氟醚处理后,小鼠饲养至3周龄,利用Morris水迷宫和Western Blot检测小鼠的神经认知功能。结果显示,A、B、C试验组逃避潜伏期与对照组相比差异不显著(P>0.05),D试验组逃避潜伏期与对照组相比差异显著(P<0.05),D组试验组中脑源性神经营养因子(BDNF)含量较对照组减少,细胞凋亡因子Caspase-3表达量较对照组增多,说明2%异氟醚吸入麻醉2 h能够显著引起小鼠神经细胞的凋亡,导致神经认知功能损伤。To investigate the influence of isoflurane on the neurocognitive function and the mechanism of impairment in 7-days old mice,45 KM mice were randomly divided into five groups:A(1.5%isoflurane exposure,1 h),B(1.5%isoflurane exposure,1.5 h),C(1.5%isoflurane exposure,2 h),D(2%isoflurane exposure,2 h)and a control group.After exposed to isoflurane,the mice were raised until 21 days old.Morris water maze and Western blot were used to test the impairment of mice’s neurocognitive function.The results showed that there were no significant differences among the escape latencies of groups A,B and C(p>0.05).However the escape latency of group D was higher than the control group(p<0.05).Furthermore,the expression of BDNF of group D were lower than the control group and the expression of Caspase-3 were higher than the control group.Therefore,exposed in 2%isoflurane for 2 h can significantly induce neuronal apoptosis,which further impaired the neurocognitive function of mice.
