摘 要:目的分析正常野生型(C57BL/6)小鼠,Toll样受体(TLR)2缺陷小鼠(TLR2-/-)诱导成中耳炎小鼠模型的听性脑干反应(ABR),分析诱导前和诱导后的ABR阈值和潜伏期,探讨中耳炎小鼠模型的听力学特点。方法将7周龄的C57小鼠和TLR2-/-小鼠各20只完全麻醉,向中耳腔鼓室注射链球菌肽聚糖多糖(PGPS)浓度为6μg/μl,每只注射10μl,3 d后内镜观察中耳炎模型建立成功。对其进行短声和短纯音ABR测试,观察两种小鼠诱导前和诱导后的ABR检测结果及各波的潜伏期。结果①诱导后的两种品系小鼠使用内镜观察到中耳炎表征。②两种品系中耳炎模型小鼠建立后ABR短声和短纯音阈值均升高,其中对短声和短纯音8 kHz最敏感,且TLR2-/-中耳炎模型小鼠的ABR反应阈值高于C57中耳炎模型小鼠,同样是对低频刺激最为敏感。③麻醉情况下,反应阈敏感的短声(click)在80 dB SPL强度下,TLR2-/-中耳炎模型小鼠的Ⅰ波、Ⅱ波、Ⅳ波潜伏期均较诱导前延长,且有统计学意义,C57中耳炎模型小鼠的Ⅰ波、Ⅱ波、Ⅳ波潜伏期也较诱导前明显延长,Ⅰ波、Ⅱ波差异有统计学意义(P<0.05)。结论利用PGPS鼓室注射成功构建中耳炎小鼠模型,C57小鼠和TLR2-/-中耳炎模型小鼠的ABR反应阈值均较正常小鼠明显升高,同时TLR2-/-中耳炎模型小鼠的ABR反应阈值明显高于C57中耳炎模型小鼠,且对低频刺激最为敏感,TLR2因子的缺失可能对听力水平的降低造成影响;同样在低频短声刺激下,两种中耳炎模型小鼠的Ⅰ波、Ⅱ波潜伏期均较正常小鼠延长。Objective To analyze auditory brainstem response(ABR)of normal wild-type mice(C57 BL/6)and toll-like receptor(TLR)2 defects(TLR2-/-)mice which were induced into otitis media model,detect ABR threshold and the incubation period before and after induction,and study the audiological characteristics of otitis media model mice.Methods All 20 of 7-week-old C57 mice and TLR2-/-mice were completely anesthetized,and injected streptococcus peptides polysaccharide(PGPS)into the middle ear drum chamber,and endoscopic observation was used to identify successful model after three days of injection.The ABR detection results and the incubation period of waves were observed in click and tone burst examination before and after induction.Results Characterization of otitis were observed by endoscopy in the two strains of mice.After model establishment,threshold of ABR click and tone burst were higher than those of before model establishment,otitis media model mice were most sensitive to click and 8 kHz tone burst.Threshold of ABR in TLR2-/-mice model was higher than that of C57 mice.Under anesthesia condition,incubation period of waveⅠ,waveⅡand waveⅣwere significantly extended in TLR2-/-mice model under the strength of 80 dB SPL click with sensitive response threshold,the differences were statistically significant.Incubation period of waveⅠ,waveⅡand waveⅣwere significantly extended in C57 mice model,the differences were statistically significant in waveⅠand waveⅡ.Conclusions Injection PGPS into the middle ear drum chamber can make otitis media model successfully.Threshold of ABR in C57 and TLR2-/-otitis media model mice are higher than those of normal mice,and threshold of TLR2-/-mice is higher than that of C57 mice,and TLR2-/-otitis media model mice is most sensitive to low frequency stimulation.Lack of TLR2 factor might decrease the hearing level.Under low frequency click stimulation,incubation period of waveⅠand waveⅡare extended in two kinds of mice than those of normal mice.
