摘　　要：目的研究橙皮素对SW480结肠癌细胞的诱导凋亡作用及其机制。方法将SW480结肠癌细胞分成4组:空白组(未给药)和低、中、高3个剂量实验组(橙皮素100,200,400μmol·L^-1),体外干预24,36和48 h。以CCK-8法检测细胞活力,以比色法检测半胱天冬酶3(Caspase3)凋亡蛋白酶活力,用实时荧光定量PCR检测p53基因的表达水平。结果干预24 h后,空白组和低、中、高3个剂量实验组的细胞活力(OD值)分别为1.00±0.00,0.83±0.17,0.75±0.15和0.58±0.12;空白组和低、中、高3个剂量实验组的Caspase3凋亡蛋白酶活力(OD值)分别为1.00±0.00,1.83±0.34,2.14±0.38和2.55±0.28;空白组和低、中、高3个剂量实验组的p53基因表达水平分别为1.00±0.00,2.12±0.34,2.86±0.29和3.39±0.51;低、中、高3个剂量实验组与空白组相比,上述指标的差异均有统计学意义(P<0.05,P<0.01)。结论橙皮素能够有效诱导SW480结肠癌细胞进入细胞凋亡进程。Objective To study the effect of hesperetin on apoptosis of SW480 colon cancer cells and its mechanism.Methods SW480 colon cancer cells were divided into 4 groups:blank group(not administered)and low,medium and high dose experimental groups(hesperetin 100,200,400μmol·L^-1),in vitro intervention 24,36 and 48 h.The cell viability was detected by CCK-8 method.The activity of Caspase3 was detected by colorimetric assay.The expression of p53 gene was detected by real time quantitative-PCR.Results After 24 h of intervention,the cell viability(OD values)of blank group and low,medium and high dose experimental groups were 1.00±0.00,0.83±0.17,0.75±0.15 and 0.58±0.12,respectively;the Caspase3 apoptosis protease activity(OD value)of blank group and three doses experimental groups were 1.00±0.00,1.83±0.34,2.14±0.38 and 2.55±0.28,respectively;the expression gene of the p53 in the blank group and the three doses experimental groups were 1.00±0.00,2.12±0.34,2.86±0.29 and 3.39±0.51,respectively.Comparison between the three high-dose experimental groups and blank group,the difference of the factors were significantly(P<0.05,P<0.01).Conclusion Hesperetin can effectively induce the apoptosis of SW480 colon cancer cells.
