摘 要:目的基于Toll样受体(TLR)/核因子-κB(TLR/NF-κB)信号通路探讨微小RNA-146a(miR-146a)在急性脑缺血大鼠脑损伤的作用。方法按照体重将大鼠随机分为3组:假手术组,模型组和实验组,每组20只。用双侧颈总动脉钳夹构建急性脑缺大鼠为模型。建模后,实验组尾部静脉注射8.0 mg·kg^-1miR-146a激动剂。用酶联免疫吸附实验检测大鼠血清肿瘤坏死因子α(TNF-α)、白细胞介素1(IL^-1)、白细胞介素6(IL-6)和核因子-κB(NF-κB)含量,用蛋白免疫印迹检测核因子/k基因结合核因子抗体(NF-κB p65)、肿瘤坏死因子受体相关因子6(TRAF-6)、白细胞介素1受体相关激酶-1(IRAK-1)蛋白的表达。结果假手术组、模型组和实验组的IL^-1分别为(351.87±21.21),(819.61±22.29)和(403.11±22.21)pg·m L^-1;这3组的IL-6分别为(187.21±12.12),(591.41±12.29)和(342.22±12.22)pg·m L^-1;这3组的TNF-α分别为(153.85±12.56),(481.45±12.56)和(243.71±12.89)pg·m L^-1;这3组的NF-κB分别为(17.15±1.21),(58.87±1.24)和(39.21±1.27)pg·m L^-1;这3组的NF-κB p65相对表达量分别为1.02±0.08,3.43±0.19和1.87±0.14;这3组的IRAK-1相对表达量分别为0.97±0.09,1.78±0.12和1.34±0.13;这3组的TRAF-6相对表达量分别为1.02±0.11,2.41±0.16和1.28±0.13。上述指标:模型组与假手术组相比,差异均统计学意义(均P<0.05);实验组与模型组比较,差异均统计学意义(均P<0.05)。结论miR-146a激动剂可以通过TLR/NF-κB通路,参与对急性脑缺血大鼠脑损伤功能的保护。Objective To investigate the role of miR-146a in brain injury rats with acute cerebral ischemia based on Toll-like receptor/nucler factor-Kappa B(TLR/NF-κB)signaling pathway.Methods Rats were randomly divided into 3 groups according to body weight:sham operation group,model group and experimental group,20 rats in each group.Acute cerebral ischemia rats were established by bilateral common carotid artery clamps as models.The experimental group injected 8.0 mg·kg-1 miR-146a agonist intravenously into the tail after modeling.The changes of serum tumor necrosis factorα(TNF-α),interleukin-1(IL-1),interleukin-6(IL-6)and nuclear factorκB(NF-κB)contents were detected by enzyme linked immunosorbent assay.The expression of nuclear factor/k gene binding nuclear factor antibody(NF-κB p65),tumor necrosis factor receptor related factor 6(TRAF-6),interleukin 1 receptor related kinase-1(IRAK-1)proteins were detected by Western blot.Results IL-1 contents in sham operation group,model group and experimental group were(351.87±21.21),(819.61±22.29)and(403.11±22.21)pg·mL-1,respectively;IL-6 contents in the three groups were(187.21±12.12),(591.41±12.29)and(342.22±12.22)pg·mL-1,respectively;TNF-αcontents in the three groups were(153.85±12.56),(481.45±12.56)and(243.71±12.89)pg·mL-1,respectively;NF-κB contents in the three groups were(17.15±1.21),(58.87±1.24)and(39.21±1.27)pg·mL-1,respectively;the relative expression of NF-κB p65 contents in the three groups were 1.02±0.08,3.43±0.19 and 1.87±0.14,respectively;the relative expression of IRAK-1 in the three groups were 0.97±0.09,1.78±0.12 and 1.34±0.13,respectively,and the relative expression of TRAF-6 in the three groups were 1.02±0.11,2.41±0.16 and 1.28±0.13,respectively.Comparison between model group and sham operation group,the difference of the factors were significantly(all P<0.05);Comparison between experimental group and model group,the difference of the factors were significantly(all P<0.05).Conclusion miR-146a agonists can participate in the pro
