摘  要:目的了解海口市农贸市场家禽中艾伯特埃希菌的带染情况,为艾伯特埃希菌检测方法的有效性和可行性提供证据。方法采集海口市不同农贸市场市售鸡肠、鸭肠内容物经EC肉汤增菌后,PCR对其进行eae基因检测,阳性增菌液接种麦康凯琼脂,挑取eae阳性、不发酵乳糖菌落进行艾伯特埃希菌筛查后经三重PCR(clpX、lysP和mdh)及多位点序列分型(MLST)鉴定。结果本研究共采集了221份样品(鸡肠及其内容物133份,鸭肠及其内容物88份),从221份鸡肠、鸭肠内容物中检出11株疑似艾伯特埃希菌,经eae基因检测、生化鉴定、MLST聚类分析,结果表明该11株菌均为不发酵乳糖、木糖,无动力,eae基因和三重PCR为阳性,MLST聚类分析与艾伯特埃希菌标准菌株具有高度亲缘性,均鉴定为艾伯特埃希菌。结论海口市地区不同农贸市场家禽中艾伯特埃希菌存在不同程度的带染,且艾伯特埃希菌检测技术有效可行,具有推广应用价值。Objective To investigate the prevalence of Escherichia albertii in Haikou's farm market and provide basis to prove the method efficiency and feasibility of Escherichia albertii detection.Methods Enriched with EC broth,the retail duck and chicken intestines collected from different farm markets in Haikou were used for detecting eae gene by PCR.The positive EC broth in PCR detection of eae gene was inoculated in MacConkey.Those eae-positive lactose non-fermenting isolates were further classified as Escherichia albertii through triple PCR(clpXf lysP and mdh)and MLST(multilocus sequence typing).Results This study collected 221 samples including 133 chicken intestines and 88 duck intestines.Among 211 samples,there were 11 suspected Escherichia albertii detected.They were all non-fermenting lactose,xylose and defunct strains,whose results were eae-positive in triple PCR.After MLST cluster analysis,the 11 strains were highly related to the reference strain of Escherichia albertii and could be confirmed as Escherichia albertii.Conclusion This study indicated that Escherichia albertii existed in varying degrees in different farm markets in Haikou.Furthermore,the technology of Escherichia albertii detection was effective and feasible,which has applicable value.
