摘  要:目的以梓醇、地黄苷A、地黄苷D、益母草苷、毛蕊花糖苷5种地黄苷类成分含量为指标优选地黄苷类提取物提取工艺;测定其对甲亢阴虚大鼠环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)及三碘甲腺原氨酸(T3)、甲状腺素(T4)的影响。方法采用HPLC-PDA法同时测定地黄中5种苷类成分含量,以正交试验法筛选最优提取工艺;采用甲状腺片灌胃建立阴虚模型动物,检测地黄苷类提取物对阴虚模型大鼠血清中T3、T4的影响及血浆中cAMP、cGMP、cAMP/cGMP的影响。结果地黄苷类提取物最佳工艺为A2B2C3D1,即加8倍量50%乙醇65℃提取3次,第1次,第2次1.5h,第3次1h。该提取物显著降低阴虚大鼠血清中T3、T4含量及降低血浆中cAMP、cAMP/cGMP值。结论该制备工艺稳定可控,地黄苷类提取物平均提取率33%,5种苷类成分提取量达6mg/g以上;针对甲亢阴虚模型大鼠,可通过调节血清中T3、T4的含量和血浆中cAMP、cAMP/cGMP而达到滋阴清热作用。Objective Optimization the extraction process of Rehmannin extracts by the contents of catalpol,Rehmannin A,Rehmannin D,Leonurus glycoside and Mulberry glycoside being used as index components,and determine its effects on cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP),triiodothyronine(T3)and thyroxine(T4)in Hyperthyroidism Rats with Yin deficiency.Methods The contents of five glycosides in Radix Rehmanniae were simultaneously determined by HPLC-PDA,and the optimum extraction process was screened by orthogonal test design.The effects of Rehmannin extracts on T3 and T4 in serum and cAMP,cGMP and cAMP/cGMP in plasma of Yin deficiency model rats were detected by intragastric administration of thyroid tablets.Results The optimum extraction process of Rehmannia glutinoside was A2B2C3D1,which was extracted three times at 65℃with 8 times of 50%ethanol,1.5 hr for the first and second time and 1 hr for the third time.The extract can significantly reduce the content of T3 and T4 in serum and the value of cAMP and cAMP/cGMP in plasma of Yin deficiency rats.Conclusion The extraction process is stable and controllable.The average extraction rate of Rehmannia glycosides extracts is 33%.The extraction amount of five glycosides is more than 6 mg/g.For hyperthyroidism Yin deficiency model rats,the effect of nourishing Yin can be achieved by regulating the content of T3 and T4 in serum and cAMP and cAMP/cGMP in plasma.
