摘 要:目的:探讨研究骨碎补有效部位对Notch通路下成骨细胞增殖及胫骨牵张成骨效能的影响。方法:选取清洁级SD雄性大鼠50只,采集大鼠成骨细胞,使用骨碎补总黄酮高、中、低剂量培养,采用MTT比色法检测细胞增殖;选取30只大鼠造模,将大鼠随机分为3组:全程用药组、牵张期用药组和牵张矿化期用药组,每组10只大鼠,记录牵张效果、影像学评分和骨密度,检测大鼠骨髓基质细胞Hes-1、Notch-1mRNA及蛋白表达水平。结果:高、中、低剂量组大鼠骨髓基质细胞Hes-1、Notch-1mRNA和蛋白表达水平均低于对照组,差异有统计学意义(P<0.05),高剂量组大鼠骨髓基质细胞Hes-1、Notch-1mRNA和蛋白表达水平均低于中、低剂量组,差异有统计学意义(P<0.05);高、中、低剂量组TGFβ-1、BMP-2、ALP和光密度值均高于对照组,差异有统计学意义(P<0.05),高、中剂量组BMP-2、ALP和光密度值高于低剂量组,差异有统计学意义(P<0.05);A组大鼠X线评分和骨密度值均高于B、C组,差异有统计学意义(P<0.05),B、C组间X线评分和骨密度值差异无统计学意义(P>0.05)。结论:骨碎补总黄酮可以通过抑制Notch通路,促进成骨细胞增殖,明显提高胫骨牵张成骨区成骨质量。Objective:To investigate the effect of total flavonoids of Rhizoma Drynariae on osteoblast proliferation and tibia distraction osteogenesis for Notch pathway. Methods:Fifty male SD rats of clean grade were selected to collect rat osteoblasts, and the proliferation of osteoblasts was detected by MTT colorimetric assay in high, medium and low doses of total flavonoids of Rhizoma Drynariae. Thirty rats were selected to establish models and randomly divided into three groups:whole course medication group, distraction medication group and distraction mineralization medication group. Ten rats in each group were recorded distraction effect, imaging score and bone mineral density. The expression levels of Hes-1 and Notch-1 in rat bone marrow stromal cells were detected. Results:The expression levels of Hes-1 and Notch-1 mRNA and protein in bone marrow stromal cells of rats in high, medium and low dose groups were lower than those in the controlled group(P<0.05). The high dose group of rat bone marrow stromal cells Hes-1, Notch-1 mRNA and protein expression levels were lower than the middle and low dose groups, the difference was statistically significant(P<0.05). High, medium and low dose groups TGFβ-1, BMP-2, ALP and optical density values. The difference was statistically significant(P<0.05). The BMP-2, ALP and optical density values in the high and middle dose groups were higher than those in the low dose group(P<0.05). The X-ray score and bone mineral density were higher than those in group B and C. The difference was statistically significant(P<0.05). There was no significant difference between X-ray score and bone mineral density between groups B and C(P>0.05). Conclusion:The total flavonoids of Rhizoma Drynariae can promote the proliferation of osteoblasts by inhibiting the Notch pathway, and significantly improve the osteogenesis quality of the tibia distraction osteogenesis.
