摘 要:利用RT-PCR结合RACE技术获得了苹果茎痘病毒(Apple stem pitting virus,ASPV)内蒙古‘金红’苹果分离物(ASPV-NM)全长基因组序列(登录号:MK239268)。该分离物基因组除去Poly A尾共有9286个核苷酸,与17个已经报道的ASPV分离物全长基因组序列核酸一致性为70.6%~79.2%;基于全长基因组、RNA依赖的RNA聚合酶(RNA-dependent RNA polymerase,RdRp)基因和外壳蛋白(Coat protein,CP)基因分别将分离物ASPV-NM划分到组Ⅲ、组Ⅲ和组Ⅰ;在ASPV-NM基因组中检测到1个显著的重组事件;将ASPV-NM摩擦接种到西方烟37B上,可引起显著的褪绿黄化症状,利用透射电子显微镜可以观察到典型的病毒粒子。One Apple spot pitting virus(ASPV)isolate NM was collected from an infected‘Jinhong’apple in Hohhot,Inner Mongolia,China,and its complete genome sequence was determined by RT-PCR and RACE techniques.The Genbank accession No.is MK239268.The single-stranded positive RNA genome of ASPV-NM had 9286 nucleotides,excluding the poly A tail,and its shared sequence identities with the 17 published ASPV genomes in NCBI were from 70.6% to 79.2%.The phylogenetic trees based on complete genome,RNA-dependent RNA polymerase(RdRp)and Coat protein(CP)sequences showed ASPV-NM clustered into Group Ⅲ,Ⅲ andⅠ,respectively.One clear recombination event was identified in the isolate ASPV-NM.The ASPV-NM was sap-inoculated into Nicotiana occidentalis 37B,causing typical symptoms of yellow spots and chlorosis,and the infection was further confirmed by the transmission electron microscope.
