摘  要:旨在明确内蒙古地区猪传染性胃肠炎流行毒株S基因遗传进化关系,为该病的防控提供依据。对内蒙古赤峰市采集的腹泻样品进行TGEV分离鉴定,成功分离到1株TGEV,将其命名为TGEV-CF(NMG)株。对TGEV-CF(NMG)株进行S基因的克隆、测序并与国内外已报道的25条基因序列进行比对和遗传进化分析。遗传进化分析结果表明,TGEV-CF(NMG)株与中国的attenuated H株和H16株亲缘关系最近,位于同一分支上。核苷酸序列比对结果显示,TGEV-CF(NMG)株与其他25株TGEV的同源性在95.1%~99.2%之间,其中与中国H16株同源性最高,为99.2%。表明分离株TGEV-CF(NMG)的S基因保守性较高,可作为制备TGEV疫苗的重要候选毒株。The aim of this study was to clarify the genetic and evolutionary relationship of porcine transmissible gastroenteritis virus S gene in Inner Mongolia,and to provide a basis for the prevention and control of this disease.The diarrhea samples were separated and identified by TGEV collected from Chifeng City of Inner Mongolia,and a TGEV strain was successfully isolated and named TGEV-CF(NMG).S gene of TGEV-CF(NMG)strain was cloned and analyzed along with the sequence alignment and phylogenetic trees of 25 S gene sequences from other domestic and international.phylogenetic analysis indicated that the relatives of TGEV-CF(NMG)strain were closest to attenuated H strain and H16 strain from China,which were located in the same branch.Nucleotide sequence alignment showed that the homology of TGEV-CF(NMG)strain with other 25 TGEV strains was between 95.1%and 99.2%,among which the homology with H16 strain in China was the highest(99.2%).The results showed that the S gene of TGEV-CF(NMG)strain was highly conserved and could be used as an important candidate strain for the preparation of TGEV vaccine.
