高效液相色谱同时测定桂生汤中毛蕊花糖苷和桂皮醛的含量

摘　　要：目的建立高效液相色谱法同时测定桂生汤中毛蕊花糖苷和桂皮醛的含量。方法样品经微孔滤膜过滤后直接进样,色谱柱为Agilent ZORBAX Eclipse XDB-C18柱(4.6 mm×250 mm,5μm),以乙腈(A)-0.05%磷酸(B)为流动相梯度洗脱,流速1.0 mL·min^-1,检测波长290 nm,柱温25℃。结果毛蕊花糖苷线性范围为0.010~0.242 mg·mL^-1,桂皮醛线性范围为0.020~0.494 mg·mL^-1,分离度良好。结论该方法简便可靠,专属性强,为桂生汤的进一步制剂研究提供了含量测定方法。Objective To simultaneously determine verbascoside and cinnamaldehyde in Guisheng decoction by HPLC.Methods Samples were directly injected after microporous membrane filtration.The chromatographic column was Agilent ZORBAX Eclipse XDB-C18(4.6 mm×250 mm,5μm).Acetonitrile(A)-0.1%phosphoric acid(B)was used as the mobile phase with gradient elution.The flow rate was 1.0 mL·min^-1.The detection wavelength was 290 nm and the column temperature was 25℃.Results The linear range of verbascoside and cinnamaldehyde was 0.010-0.242 mg·mL^-1 and 0.020-0.494 mg·mL^-1.The two substances were well separated.Conclusion The method is simple,reliable and specific,which is suitable for the determination of Guisheng decoction.
