摘  要:背根神经节细胞(DRG)是外周神经系统中感受、传导痛觉的重要细胞,多聚赖氨酸是神经细胞体外培养常用的细胞外基质(ECM),但不同类型、不同浓度的多聚赖氨酸对背根神经节细胞生长分化的具体影响目前仍未阐明。目的:本项目旨在探讨不同类型、浓度、分子量的多聚赖氨酸对大鼠背根神经节细胞生长分化的影响。方法:分别从时间梯度、浓度梯度以及不同分子量这三个方面研究D型及L型多聚赖氨酸对大鼠背根神经节细胞分化率及平均突起长度的影响。结果:实验结果表明,正常工作浓度下用L型多聚赖氨酸作为基底材料培养细胞48 h时对细胞突起长度有一定的促进作用;当D型、L型多聚赖氨酸的浓度分别降低至5 g/ml、20 g/ml时,细胞在L型多聚赖氨酸包被的玻片上分化更好;而普通分子量(3-7 W)或高分子量(>30 W)的D型多聚赖氨酸对背根神经节细胞的生长分化无显著影响。Dorsal root ganglion cells(DRG)are important cells in the peripheral nervous system to sense and transmit pain.Polylysine is a common extracellular matrix(ECM)for nerve cells cultured in vitro.However,the specific effects of different types and concentrations of polylysine on the growth and differentiation of dorsal root ganglion cells have not yet been clarified.Objective:To investigate the effects of polylysine of different types,concentration and molecular weight on the growth and differentiation of rat dorsal root ganglion cells.Methods:The effects of D-type and L-type polylysine on the differentiation rate and average process length of rat dorsal root ganglion cells were studied from three aspects:time gradient,concentration gradient and molecular weight.Results:The results showed that L-polylysine could promote the length of cell processes at 48 hours in normal working concentration.When the concentration of D-polylysine and L-polylysine decreased to 5 ug/ml and 20 ug/ml respectively,the cells differentiated better on L-polylysine-coated slide,while the normal molecular weight(3-7 W)or high molecular weight(>3 ug/ml)were better.0 W)D-polylysine had no significant effect on the growth and differentiation of dorsal root ganglion cells.
