摘  要:几丁质是自然界广泛存在的第二大氨基多糖,在几丁质的合成途径中有许多酶的参与,其中磷酸乙酰氨基葡萄糖变位酶(Phosphoacetylglucosamine mutase,PAGM)是关键酶之一。以飞蝗为研究对象,对磷酸乙酰氨基葡萄糖变位酶基因PAGM的表达及功能进行了研究。对飞蝗转录组数据库进行搜索,得到PAGM基因全长cDNA序列,将其命名为LmPAGM(GenBank登录号:MK568624),其中开放阅读框长度1 491 bp,编码497个氨基酸。通过Real-time PCR分析该基因在五龄若虫不同组织部位和不同天数的表达,结果表明LmPAGM在体壁、前肠、中肠、后肠、胃盲囊、马氏管、脂肪体和翅中均衡表达,并且在五龄若虫第8天表达显著上升。利用RNAi对LmPAGM的生物学功能进行探讨,注射LmPAGM的dsRNA至五龄若虫第2天的飞蝗,注射dsLmPAGM组中LmPAGM的表达较对照组显著下调,并且约30%的飞蝗无法成功羽化为成虫。Chitin is the second most abundant biological polymer found in nature. Many enzymes are involved in the synthesis of chitin, and phosphoacetylglucosamine mutase(PAGM) is one of the key enzymes.The expression and function of phosphoglucose mutase gene PAGM in Locusta migratoria were studied. By searching the locust transcriptome database, the full-length cDNA sequence of PAGM gene was obtained, named LmPAGM(GenBank accession number MK568624). It contained a 1491-nucleotide open reading frame(ORF) encoding 497 amino acid residues. Real-time PCR was used to analyze the expression of LmPAGM in different tissues and developmental days of 5 th instar nymphs. The results showed that LmPAGM was expressed in all the tested tissues including integument, foregut, midgut, hindgut, gastric caecum, malpighian tubule, fat body and wings. The expression cnincreased significantly on the 8 th day of the 5 th instar nymphs. The biological function of LmPAGM was explored by RNAi. The dsRNA of LmPAGM was injected into the 2 nd day of the 5 th instar nymphs. Compared to the control, the expression of LmPAGM was significantly down-regulated in dsLmPAGM injected insects, and about 30% of locusts could not successfully molt into adults.
